基于多技术指纹图谱的金银花产地加工温度与成分变化相关性研究 被引量:2

Study on the correlation between temperature and content of Lonicerae japonicae Flos during processing

摘要 目的建立多技术指纹图谱及化学计量学分析模型,研究金银花产地加工过程中温度与成分变化的相关性规律。方法采用Waters e2695-2489高效液相系统,Symmetry C_(18)色谱柱(4.6 mm×250 mm×5μm),乙腈-0.2%甲酸水溶液梯度洗脱建立HPLC指纹图谱;采用Bruker INVENIO-HYPERION 2000傅里叶变换红外显微系统,扫描4000~400 cm;波段建立Mid-IR指纹图谱;采用中药色谱指纹图谱相似度评价系统(2012.130723版)、OMNIC 9.2.106以及SIMCA-p 14.1等软件,进行计量学分析及模型建立。结果分别建立了金银花样品HPLC及MIR指纹图谱及对照图谱,标示出16个HPLC共有峰、10个MIR特征峰;利用多元回归分析MIR指纹图谱与HPLC指纹图谱数据建立了光谱与含量的回归模型,进一步揭示了金银花产地加工过程中温度与成分变化相关性关系。结论HPLC与MIR指纹图谱技术可以多维度地呈现样本的指纹特性,结合计量学分析可应用于金银花产地加工工艺优化、全过程控制及系统性评价,同时为中药品质快速分析检测技术的开发提供思路与方法。 Objective To establish a multi-technical fingerprint and chemometrics analysis model to study the correlation between temperature and content in the producing area and the processing of L.japonicae Flos.Methods The HPLC fingerprint was established using Waters e2695-2489 HPLC system;The Mid-IR fingerprint is established using the Bruker INVENIO-HYPERION 2000 Fourier transform infrared microscopy system;The analysis method comprehensively utilizes Chinese Medicine Chromatographic Fingerprint Similarity Evaluation System(2012.130723 Edition),OMNIC 9.2.106 and SIMCA-p 14.1 to conduct multiple data analysis.Results HPLC and MIR fingerprints and control chromatograms of 10 batches of L.japonicae Flos were established respectively,and 16 common HPLC peaks and 10 MIR characteristic peaks were identified.Using OPLS to analyze the data of MIR and HPLC fingerprint and a regression model of spectral data and component content was established.Conclusion The evaluation model established by combining the multivariate data analysis method can be used for the systematic evaluation of the quality of L.japonicae Flos.
