模拟高温胁迫下玉米自交系群体花粉活力的快速测定及评估 被引量:2

Rapid Determination and Estimation of Pollen Viability under Simulated High Temperature Stress in Maize Inbred Population

摘要 基于阻抗流式细胞仪、雄穗水培及PCR仪模拟高温胁迫,建立花粉活力的快速测定体系,测定玉米自交系群体(227份)在常温(25℃)及模拟高温胁迫下(40℃/30 min、50 min、70 min)的花粉活力值,利用BLUE模型估算综合花粉活力值,用于筛选耐高温型玉米自交系。在常温下,花粉活力值在玉米自交系群体中变异丰富。随着模拟高温胁迫时间的延长,自交系间的花粉活力值存在不同程度的下降,与果穗产量的相关系数逐渐减小,表明玉米自交系的花粉活力对高温胁迫的耐受性在群体中存在差异。利用综合花粉活力值,共筛选到耐高温型玉米自交系15份,它们的花粉活力均对高温胁迫具有较高的耐受性。 Based on impedance flow cytometer,water culture and simulated high-temperature stress by PCR,the rapid determination system of pollen viability(PV)was established and PV of 227 maize inbred lines were deter⁃mined at room temperature(25℃)and simulated high-temperature(40℃/30 min,50 min,70 min),which were used in estimation of comprehensive pollen viability(CPV)by BLUE.At room temperature,variants of PV are abundant and follow normal distribution in maize inbred population.With simulated high-temperature stress,PV exhibited different degree of decline and correlation coefficients with yield of ear was also decreased,which indicated the tol⁃erance of PV to simulated high-temperature stress is significant difference in maize inbred population.A total of 15 maize inbred lines with heat-resistance was obtained by CPV,present high resistance to simulated high tempera⁃ture.The research lay the foundation for rapid,quantitative estimation of pollen viability in this paper.
