青海省野生中国沙棘根际解磷菌的初步研究

Preliminary Study on Phosphate Solubilizing Bacteria in the Rhizosphere of Wild Chinese Seabuckthorn in Qinghai Province

摘要 为探究青海省野生中国沙棘根际土中是否存在原位土著高效解磷菌,本研究从青海省西宁市湟中区野生中国沙棘根际土中进行分离纯化菌株,纯化获得72株待测解无机磷菌和24株待测解有机磷菌,通过初筛和复筛进行功能分析,确定解磷能力强弱。初筛结果显示有18株解无机磷菌株和12株解有机磷菌株。复筛结果显示:(1)解无机磷菌株WP-4(1d)菌发酵液总磷含量略高于对照,为4.40mg/L;随着培养时间的延长,所有菌株低于对照,初筛获得的菌株解无机磷能力相对较差;(2)解有机磷菌株YP-7(1d)和YP-10(1d)菌发酵液中总磷含量高于对照,分别为5.33mg/L和4.22mg/L,相对较好;其他菌株及其它天数的菌株总磷含量低于对照,故解磷能力相对较差。综上,对于此地区解磷菌初步研究的结论为其解磷活性较低。 In order to explore whether there are in situ indigenous high-efficiency phosphorus-solubilizing bacteria in the rhizosphere soil of wild Chinese sea buckthorn in Qinghai Province,in this study,strains were isolated and purified from the rhizosphere soil of wild Hippophae rhamnoides L.in Huangzhong District,Xining City,Qinghai Province,72 strains of inorganic phosphorus-dissolving bacteria and 24 organic phosphorus-dissolving bacteria were obtained through purification,and the ability of phosphorus-dissolving ability was determined by primary screening and secondary screening for ability analysis.The preliminary screening results showed that there were 18 strains of inorganic phosphorus-solving strains and 12 strains of organic phosphorus-solving strains.The results of the re-screening show:(1)The total phosphorus content of the fermentation broth of the inorganic phosphorus-solving strain WP-4(1d)was slightly higher than that of the control,which was 4.40mg/L;with the extension of the culture time,all the strains were lower than the control,the strains obtained from the primary screening have relatively poor ability to dissolve inorganic phosphorus;(2)The total phosphorus content in the fermentation broth of the organophosphorus-solubilizing strains YP-7(1d)and YP-10(1d)was higher than that of the control,which were 5.33mg/L and 4.22mg/L,respectively,which were relatively good;other strains and The total phosphorus content of the strains on other days was lower than that of the control,so the ability to dissolve phosphorus was relatively poor.To sum up,the preliminary study on phosphate-solubilizing bacteria in this area concluded that its phosphate-solubilizing activity was low.
