摘要
目的 在胆管癌(CCA)中用铁死亡相关长链非编码RNA(lncRNA)建立有较好的预测能力的预后模型。方法 从TCGA数据集下载CCA的转录组RNA-seq数据和相应的临床数据。采用Pearson相关法筛选与铁死亡相关的差异表达的lncRNAs。单因素Cox回归分析用来筛选与预后相关的lncRNAs。通过多因素回归分析构建预后模型,采用受试者工作特征曲线(ROC)下面积和Kaplan-Meier曲线对模型进行评价。结果 筛选出3个铁死亡相关的差异表达lncRNAs:LINC01714、LINC02997和LINC01621。基于这3个lncRNAs构建的预后模型,预测1年、3年、5年生存率的ROC曲线下面积分别为0.81、0.81和0.82。结论 本研究基于铁死亡相关lncRNAs建立了一个预后模型预测CCA患者的生存概率,挖掘了有潜力作为CCA患者预后标志物的lncRNAs。
Objective To develope a novel ferroptosis-related long non-coding RNAs(lncRNAs)prognostic model for cholangiocarcinoma(CCA)with high predictive accuracy.Methods The transcriptome RNA-seq data and corresponding clinical data of CCA were downloaded from the TCGA dataset.Pearson correlation was used to mine ferroptosis-related differential lncRNAs.Univariate Cox regression analysis was performed to screen prognosis related lncRNAs.Using multivariate Cox regression analysis to develop aprognostic model.The model was evaluated using the area under the receiver operating characteristic curve(ROC)and Kaplan-Meier curve.Results Three ferroptosis-related lncRNAs were screened.A ferroptosis-related lncRNAs prognostic model was constructed,which included LINC01714,LINC02997and LINC01621.The area under the ROC cure for the 1-year,3-years,and 5-years survival rate were 0.81,0.81and 0.82.Conclusion A prognostic model based on ferroptosis-related lncRNAs was created to predict the survival probability of CCA patients.Our data suggested that the prognostic lncRNAs could serve as valuable prognostic markers for CCA patients.
