摘要
目的 探讨希瓦氏菌临床株HDC555对碳青霉烯类耐药的分子机制。方法 希瓦氏菌临床株的鉴定和药敏使用VITEK 2Compact自动仪;碳青霉烯酶的筛选通过免疫金标法;S1核酸酶切脉冲场凝胶电泳(S1-PFGE)检测菌株携带的质粒。基于二代Illumina和三代Nanopore技术获取菌株的全基因组序列,并用一系列软件分析菌株的生物学信息。结果 免疫金标法显示希瓦氏菌HDC555产OXA-48样和NDM酶。高通量测序数据经分析(ANI)表明希瓦氏菌HDC555株是厦门希瓦氏菌,且该株菌携带位于染色体上的blaOXA-199基因和位于质粒上的blaNDM-1基因。结论 厦门希瓦氏菌临床株HDC555对碳青酶烯类耐药的分子机制是因为该菌携带了blaOXA-199和blaNDM-1碳青霉烯酶基因。
Objective To explore molecular mechanism of a clinical carbapenem-resistant shewanellaspp.HDC555strain.Methods The identification and antimicrobial susceptibility tests of the strain was carried out by VITEK-2Compact.Carbapenemases were screened by immunochromatographic assay.The plasmids were detected by S1nuclease digestion pulsed field gel electrophoresis(S1-PFGE).The whole genome sequence of the strain was obtained based on the Illumina and Nanopore technology,and the bioinformation of the strain was analyzed by a series of software.Results The immunochromatographic assay showed that Shewanellaspp.HDC555strain produced both OXA-48-like carbapenemase and NDM carbapenemase.High throughput sequencing data showed that strain HDC555 was Shewanella xiamenensis by ANI analysis,and the strain carried blaOXA-199gene on chromosome and blaNDM-1gene on plasmid.Conclusion The molecular mechanism of carbapenem resistance of clinical strain HDC555of S.xiamens is that the strain carries blaOXA-199and blaNDM-1carbapenemase genes.
