试验旨在筛选间苯三酚法和高效液相色谱法(HPLC)测定棉产品中游离棉酚含量准确快捷的方法,确定能够稳定棉粕、棉籽壳和棉蛋白中游离棉酚含量的烘干温度。通过间苯三酚法和高效液相色谱法,测定3种棉产品中的棉酚含量,对比回收率,筛选快捷准确检测棉酚含量的方法。结果显示:使用间苯三酚法测定棉蛋白、棉粕和棉籽壳中加标样游离棉酚含量均显著高于高效液相色谱法(P<0.05)。未烘干处理与65℃烘干处理下棉蛋白中游离棉酚含量差异不显著(P>0.05)。未烘干组处理下棉蛋白中的游离棉酚含量显著高于75、85、95、105℃烘干(P<0.05)。未烘干处理与65、75℃烘干处理下棉粕和棉籽壳中游离棉酚含量差异不显著(P>0.05)。未烘干组棉粕中的游离棉酚含量显著高于85、95、105℃烘干处理(P<0.05)。研究表明,间苯三酚法测定游离棉酚更快捷准确。不同烘干温度影响棉产品中游离棉酚的含量,烘干温度的增加游离棉酚含量降低,65℃烘干处理下的棉产品中游离棉酚含量稳定。
The experiment was to select an accurate and rapid method for determination of free gossypol in cotton products by phloroglucinol method and high performance liquid chromatography(HPLC) and the drying temperature,which could stabilize the free gossypol content in cottonseed meal, cottonseed hull and cottonseed protein. The content of gossypol in three kinds of cotton products was determined by phloroglucinol method and high performance liquid chromatography. The method of rapid and accurate detection of gossypol content was screened by comparing the recovery rate. The results showed that the content of standard free gossypol in gossypol, cottonseed meal and cottonseed shell determined by phloroglucinol method were significantly higher than those determined by HPLC(P<0.05). There was no significant difference in free gossypol content between untreated and 65 ℃ drying treatments(P>0.05). The content of free gossypol in gossypol without drying was significantly higher than that in gossypol without drying at 75, 85, 95 and 105 ℃(P<0.05). There was no significant difference in the content of free gossypol in cottonseed meal and cottonseed shell between untreated and 65 and 75 ℃ treatments(P>0.05). The content of free gossypol in non-drying group was significantly higher than that in 85, 95 and 105 ℃ drying treatments(P<0.05). The experiment indicates that phloroglucinol method for the determination of free gossypol faster and more accurate. Different drying temperatures affect the content of free gossypol in cotton products. With the increase of drying temperature, the content of free gossypol decreases, and the content of free gossypol in cotton products dried at 65 ℃ is stable.
