萨福克和湖羊杂交F1代与湖羊背最长肌转录组分析

Transcriptome analysis of longissimus dorsi muscle from F1 generation of Suffolk and Hu sheep crosses

摘要 试验比较分析萨福克与湖羊杂交F1代和湖羊纯繁羔羊背最长肌差异表达基因转录组,探索影响萨湖F1代和湖羊肉质的重要基因和关键代谢通路。采集10月龄萨福克与湖羊杂交F1代羔羊(SH)和湖羊纯繁羔羊(HH)背最长肌组织,利用RNA-seq技术进行基因转录组比较分析,差异表达基因qRT-PCR验证。结果显示:筛选出差异基因322个,得到GO数据库注释278个,参与生物过程(BP)、分子功能(MF)和细胞组成(CC)的分别为83、140和55个。得到KEGG数据库注释147个,参与216条通路,差异基因显著富集通路24条。通过查阅文献和对比分析,初步认为与肉质相关的通路为氨基酸的生物合成通路、甘油酯代谢通路、PPAR信号通路、MAPK信号通路和肌动蛋白细胞骨架的调节通路。筛选出与肉质相关的候选基因有LPIN1、CFL2、ENO3和AQP7。研究表明,在转录组水平上筛选出萨福克与湖羊杂交F1代、湖羊纯繁羔羊背最长肌差异表达基因322个,初步认为与肉质可能相关的通路11条,与肉质相关的候选基因4个。 In the study,transcriptome analysis of differentially expressed genes in longissimus dorsi muscle of F1 generation of Suffolk-Hu sheep and pure Hu sheep was conducted,to explore the important genes and key metabolic pathways affecting meat quality of F1 generation of Suffolk-Hu sheep and Hu sheep.The longissimus dorsi muscle tissues of 10-month-old the Suffolk and Hu sheep cross F1 generation lambs(SH)and lamb of the Hu sheep pure breeding(HH)were collected,transcriptome analysis of differentially gene expression was performed using RNA-SEQ technology,and qRT-PCR was used to verify the differentially expressed genes.The results showed that a total of 322 differential genes were screened out,278 were annotated in GO database,83 were involved in BP,140 were involved in MF and 55 were involved in CC.147 were annotated in KEGG database,and 216 pathways were involved,24 pathways were significantly enriched in differential genes.Through consulting literature and comparative analysis,it was preliminarily believed that the pathways related to meat quality included amino acid biosynthesis pathway,glycerol metabolism pathway,PPAR signaling pathway,MAPK signaling pathway and actin cytoskeleton regulation pathway.The candidate genes related to meat quality were LPIN1,CFL2,ENO3 and AQP7.The experiment indicates that at transcriptome level,322 differentially expressed genes in the longissimus dorsi muscle of F1 generation of Suffolk-Lake sheep hybrid and pure Lake sheep lamb are screened.It is preliminarily identified that 11 pathways may be related to meat quality,4 candidate genes are related to meat quality.
