血管微环境在器官再生与纤维化中的调控机制研究进展

摘要 血管是体内最大的表面之一,所有生理活动的进行都离不开血管.血管不仅能起到被动运输及屏障作用,覆盖于血管腔内部的血管内皮细胞通过分泌一些血管分泌因子(angiocrine factors)共同构成血管微环境,主动参与调节多种生理过程,例如,器官再生与纤维化,癌症的发生、发展等.器官受到损伤后,血管内皮细胞通过分泌不同的旁分泌因子,调控周围组织实质细胞,或者触发启动干细胞的增殖分化程序来调控器官的修复过程.因此结合目前单细胞测序技术以及多组学联合分析的技术手段解析血管微环境,探究维持血管稳态的因素可以为以血管为靶点的临床治疗手段提供强大的理论支持.本文主要介绍血管微环境在器官再生以及纤维化中的调控机制. Blood vessels possess a large surface area in vivo,and are indispensable for all physiological processes.Blood vessels play important roles in passive transport and barrier protection;moreover,the endothelial cells lining the vascular lumen deploy a set of angiocrine factors to establish the vascular niche and participate in many physiological processes,such as organ regeneration,fibrosis,and the development of cancer.After an organ is injured,endothelial cells guide repair by secreting different angiocrine factors,regulating parenchymal cells,or triggering the proliferation and differentiation of stem cells.Therefore,combining single-cell sequencing technology with multiomics incorporation analysis will help resolve the vascular niche and explore key target molecules for maintaining homeostasis of the vascular niche,which provides a strong background for clinical treatment of organ fibrosis.In this review,we focused on the regulatory mechanism of the vascular niche in organ regeneration and fibrosis.
