摘要
目的研究热休克蛋白(HSP)90α、HSP90β在结直肠癌组织和癌旁组织中的表达,探讨HSP90α、HSP90β与结直肠癌患者临床病理特征的关系,并分析两者的相关性。方法选取2016年1月至2020年12月山东第一医科大学第三附属医院胃肠外科收治的117例结直肠癌患者的肿瘤组织及癌旁组织,采用免疫组织化学法检测HSP90α和HSP90β的表达水平,分析二者与患者临床病理特征的关系以及二者表达的相关性。结果HSP90α在结直肠癌组织和癌旁组织中的阳性表达率分别为74.4%(87/117)、12.0%(14/117),两组差异具有统计学意义(χ^(2)=92.83,P<0.001);HSP90β在结直肠癌组织和癌旁组织中的阳性表达率分别为61.5%(72/117)、10.3%(12/117),两组差异具有统计学意义(χ^(2)=66.86,P<0.001)。HSP90α的表达与肿瘤位置(χ^(2)=8.67,P=0.003)、血管侵犯(χ^(2)=8.68,P=0.003)、淋巴结转移(χ^(2)=8.52,P=0.004)、T分期(χ^(2)=21.07,P<0.001)、N分期(χ^(2)=11.94,P=0.003)、M分期(χ^(2)=5.37,P=0.020)、病理分期(χ^(2)=25.64,P<0.001)具有相关性;HSP90β的表达与淋巴结转移(χ^(2)=4.03,P=0.045)、T分期(χ^(2)=11.09,P=0.007)、N分期(χ^(2)=6.56,P=0.038)、M分期(χ^(2)=12.43,P<0.001)、病理分期(χ^(2)=17.34,P=0.001)具有相关性。两种蛋白在结直肠癌组织中的表达呈正相关(r=0.42,P<0.001)。结论HSP90α、HSP90β在结直肠癌组织中的表达明显高于癌旁组织,并均与淋巴结转移和病理分期有关;二者表达呈正相关,可能共同参与了结直肠癌的发生发展,有望成为新的肿瘤标志物。
Objective To study the expressions of heat shock protein(HSP)90αand HSP90βin colorectal cancer and paracancer tissues,and to investigate the relationships between HSP90α,HSP90βand clinicopathological features of colorectal cancer patients,and to analyze their correlation.Methods The tumor tissues and paracancer tissues of 117 patients with colorectal cancer were selected from the Department of Gastrointestinal Surgery,Third Affiliated Hospital of Shandong First Medical University from January 2016 to December 2020.The expression levels of HSP90αand HSP90βwere detected by immunohistochemistry,and the relationships between the two proteins and clinicopathological features and the correlation of their expressions were analyzed.Results The positive expression rates of HSP90αin colorectal cancer tissues and paracancer tissues were 74.4%(87/117)and 12.0%(14/117),and there was a statistically significant difference(χ^(2)=92.83,P<0.001).The positive expression rate of HSP90βin colorectal cancer tissues and paracancer tissues was 61.5%(72/117)and 10.3%(12/117),and there was a statistically significant difference(χ^(2)=66.86,P<0.001).The expression of HSP90αwas correlated with tumor location(χ^(2)=8.67,P=0.003),vascular invasion(χ^(2)=8.68,P=0.003),lymph node metastasis(χ^(2)=8.52,P=0.004),T stage(χ^(2)=21.07,P<0.001),N stage(χ^(2)=11.94,P=0.003),M stage(χ^(2)=5.37,P=0.020),pathological stage(χ^(2)=25.64,P<0.001).The expression of HSP90βwas correlated with lymph node metastasis(χ^(2)=4.03,P=0.045),T stage(χ^(2)=11.09,P=0.007),N stage(χ^(2)=6.56,P=0.038),M stage(χ^(2)=12.43,P<0.001),pathological stage(χ^(2)=17.34,P=0.001).There was a positive correlation between the expressions of the two proteins in colorectal cancer tissues(r=0.42,P<0.001).Conclusion The expressions of HSP90αand HSP90βin colorectal cancer tissues are significantly higher than those in paracancer tissues,and they are related to lymph node metastasis and pathological stage.There is a positive correlation between the two proteins,which may be involved in the occurrence and development of colorectal cancer and are expected to become new tumor markers.
