摘要
试验旨在提取野菊茎叶多糖,并评价多糖的安全性。通过突变性检测(Ames)、小鼠骨髓嗜多染红细胞微核、小鼠精子畸形等试验考察致突变作用和一般毒性,Ames试验使用平板掺入法。结果显示:野菊茎叶多糖未诱导TA_(97)、TA_(98)、TA_(100)、TA_(102)的回变菌落数增加。雌雄鼠CP阳性对照组测试物剂量组嗜多染红细胞/正染红细胞(PCE/NCE)比值高于对照组,未见诱发小鼠骨髓嗜多染红细胞微核细胞率增高。CP阳性对照组精子畸形率极显著高于阴性对照组(P<0.01),未诱发小鼠精子畸形率增高。30 d喂养试验,低剂量组雌鼠的第3 w食物利用率显著降低(P<0.05),内脏组织病理学检查未见高剂量组的野菊茎叶多糖引起大鼠中毒性损伤改变。研究表明,野菊茎叶多糖安全无毒,无致突变作用和中毒性损伤改变。
The polysaccharide was extracted from the branches and leaves of Chrysanthemum indicum, and the safety assessment of polysaccharide was evaluated. Ames test in rats, mouse bone marrow cell micronucleus test, mouse sperm abnormality test were examined to evaluate mutagenicity and toxicities change of the polysaccharide. Ames test used the plate incorporation method. The results showed that polysaccharide from stems and leaves did not induce an increase in number of bacterial colonies of TA_(97),TA_(98),TA_(100) and TA_(102).The ratio of PCE/NCE in CP positive control group was higher than that in control group, and no increase in micronucleus rate of polychromatic erythrocytes in bone marrow was observed. The rank sum test showed that the sperm malformation rate in the CP-positive control group was extremely higher than that in negative control group(P<0.01). 30 d feeding test, the food utilization rate of female rats in low-dose group was significantly decreased at 3 w(P<0.05), and visceral histopathological examination showed no toxic damage caused by polysaccharide in high-dose group. The experiment indicates that the extracted polysaccharide of Chrysanthemum indicum is safety without toxicity and mutagenicity.
