理中汤改善抗生素相关性腹泻及对肠道菌群的干预作用研究 被引量:9

摘要 目的:研究理中汤对抗生素相关性腹泻(AAD)大鼠的干预作用及对肠道菌群的作用。方法:建立AAD大鼠模型,以双歧杆菌三联活菌胶囊为阳性药(Bif,70 mg/d),理中汤分别以3.75、7.5 g/kg(按生药量计算)给药进行干预。观察大鼠一般状况,通过120 min粪便重量、粪便含水量及粪便评分评价大鼠腹泻程度;对大鼠结肠组织进行苏木精-伊红染色(HE染色),观察其病理变化;采用16S rRNA高通量测序技术分析大鼠肠道菌群。结果:Bif给药后,大鼠腹泻程度有一定程度改善;理中汤高、低剂量治疗组大鼠粪便含水量、粪便评分及120 min粪便重量均有明显改善,其中低剂量组药效略优于高剂量组。大鼠结肠组织HE染色结果显示,理中汤高、低剂量治疗可以改善大鼠结肠炎症等病理改变。肠道菌群分析结果显示,Bif及理中汤干预后,AAD大鼠肠道菌群丰度和多样性均轻微改善,两者的差异无统计学意义(P>0.1);但理中汤在门水平上可显著恢复厚壁菌门丰度(P<0.05),在属水平上可显著恢复克雷伯菌属菌群(P<0.05)及Parasutterella属菌群(P<0.05)丰度,与AAD模型组比较,差异均有统计学意义;理中汤在属水平上可恢复拟杆菌属菌群,但与AAD模型组比较,差异无统计学意义(0.05<P<0.1)。结论:理中汤可显著改善AAD,并在一定程度上恢复肠道菌群。 OBJECTIVE:To study the intervention effects of Lizhong decoction on antibiotic-associated diarrhea(AAD),and to reveal the modulation of gut microbiota during the alleviation of AAD.METHODS:AAD rat model was established,and Bifidobacterium triple viable capsules were used as the positive control(Bif,70 mg/d).Lizhong decoction was administered at 3.75 g/kg and 7.5 g/kg(measured as the quantity of crude material).The general condition of the rats was observed,and the diarrhea degree of the rats was evaluated by 120 min fecal weight,fecal water content and fecal score.Colon tissues for histopathological examination were stained with hematoxylin and eosin(HE)staining,and 16S rRNA sequencing analysis of gut microbiota was performed.RESULTS:After administration of Bif,the degree of diarrhea in rats was alleviated.The fecal water content,fecal score and 120 min fecal weight of rats in the high-and low-dose Lizhong decoction groups showed significant improvements,with the low-dose group being slightly more effective than the high-dose group.Results of HE staining of rat colon tissue showed that the treatment of high-and low-dose Lizhong decoction could improve the pathological changes such as colon inflammation in rats.Results of gut microbiota analysis showed that after the intervention of Bif and Lizhong decoction,the abundance and diversity of gut microbiota in AAD rats were slightly improved,and there was no significant difference between them(P>0.1).However,compared with the AAD model group,Lizhong decoction significantly restored the abundance of Firmicutes at the phylum level(P<0.05),Klebsiella spp.at the genus level(P<0.05)and Parasutterella spp.at the genus level(P<0.05),the differences were statistically significant.Lizhong decoction restored the Bacteroides spp.at the genus level,yet the difference was not statistically significant compared with the AAD model group(0.05<P<0.1).CONCLUSIONS:Lizhong decoction could significantly improve AAD and positively affected the gut microbiota of rats with AAD.
