摘要
目的探讨miR-145和miR-34a在宫颈癌及癌前病变患者宫颈组织中的表达,并分析其与HPV16感染的相关性。方法选取2017年6月至2020年6月本院经宫颈活检病理学确诊的41例宫颈癌患者为A组,42例癌前病变患者为B组,40例慢性宫颈炎患者为C组。3组均行miR-145、miR-34a及HPV16感染检测,比较3组宫颈组织中miR-34a和miR-145的表达水平及HPV16感染阳性率,采用Spearman相关性分析3组宫颈组织中miR-34a和miR-145的表达与HPV16感染的相关性。结果A组宫颈组织中miR-34a和miR-145的表达水平显著低于B组和C组,B组宫颈组织中miR-34a和miR-145的表达水平显著低于C组(P<0.05)。A组HPV16感染阳性率显著低于B组、C组,B组HPV16感染阳性率显著低于C组(P<0.05)。Spearman相关性分析显示,宫颈癌及癌前病变患者组织中miR-34a和miR-145的表达与HPV16感染呈高度负相关(P<0.05),同时,宫颈癌组织中相关性更高,慢性宫颈炎患者组织中上述表达与HPV16感染无相关性。结论宫颈癌及癌前病变患者miR-145、miR-34a呈低表达,与HPV16感染显著相关,miR-145、miR-34a表达水平越低,患者HPV16感染风险越高,未来可将miR-145和miR-34a动态监测结果作为宫颈癌及癌前病变早期筛查与诊断的重要标志物。
Objective To investigate the expression of miR-145 and miR-34a in cervical tissues of patients with cervical cancer and precancerous lesions,and to analyze and their correlation with HPV16 infection.Methods From our hospital from June 2017 to June 202041 patients with cervical cancer diagnosed by science were selected as group A,42 patients with precancerous lesions were selected as group B,and 40 patients with cervicitis were selected as group C.miR-145,miR-34a and HPV16 infection were detected in all three groups,the expression levels of miR-34a and miR-145 in cervical tissue and the positive rate of HPV16 expression in the three groups of patients.Spearman correlation was used to analyze the correlation between the expression of miR-34a and miR-145 in cervical tissue of the three groups and HPV16 infection.Results The expression levels of miR-34a and miR-145 in group A were significantly lower than those of group B and group C,the expression levels of miR-34a and miR-145 in the group B was significantly lower than that of group C(P<0.05).The positive rate of HPV16 infection in group A was significantly lower than that in group B and group C,and the positive rate of HPV16 infection in group B was significantly lower than that in group C(P<0.05).Spearman correlation analysisshowed,the expressions of miR-34a and miR-145 in cervical cancer and precancerous lesions were significantly negatively correlated with HPV16 infection(P<0.05),and the correlation was higher in cervical cancer tissues and cervicitis patients.The above expressions in the tissues were not related to HPV16 infection.Conclusion Cervical cancer and precancerous lesions showed low expression of miR-145 and miR-34a,which were significantly correlated with HPV16 infection,miR-145 and miR-34a expression levels.The lower the risk of HPV16 infection,the higher the miR-145 and miR-34a dynamic monitoring results can be used as an important marker for early screening and diagnosis of cervical cancer and precancerous lesions.
