摘要
目的:建立维吾尔药材睡莲花的质量标准。方法:采用显微鉴别、TLC法鉴别;对水分、灰分、酸不溶性灰分、浸出物进行检查;采用HPLC法测定睡莲花中山柰酚-3-O-芸香糖苷的含量。色谱柱为C_(18)柱(250 mm×4.6 mm,5μm),流动相A为乙腈,流动相B为0.1%磷酸溶液,梯度洗脱;检测波长为350 nm。结果:对睡莲花药材的性状、显微特征进行了描述、TLC色谱斑点清晰,水分、灰分、酸不溶灰分的含量分别不得超过12.0%,12.0%,2.0%,浸出物不得少于30.0%,山柰酚-3-O-芸香糖苷在5.11~51.07μg/mL范围内线性关系良好(r=0.9999),平均回收率为98.0%,RSD为1.1%(n=9)。结论:本方法准确、简便、灵敏度高,能有效控制睡莲花药材的质量。
Objective To establish the quality standard of Uygur Medicine Nymphaea Candida.Methods The Nymphaea Candida was identified with the method of Microscopic identification and thin-layer chromatography(TLC),The content of moisture,total ash,acid-insoluble ash and extract were determined.and the HPLC method was used to determine the contents of kaempferol-3-O-rutinoside in Nymphaea Candida with C_(18)column(250 mm×4.6 mm,5μm).The mobile phase A was acetonitrile,The mobile phase B was 0.1%phosphoric acid.gradient elution and the detection wavelength was 350 nm.Results The character and microscopic characteristics of Cymbopogon caesius were described.TLC identification method was established and the spots were clear and well-separated.The content of moisture could not more than 12.0%,total ash could not be more than 12.0%,acid-insoluble ash could not be more than 2.0%,and extract could not be less than 30.0%,The calibration curve of kaempferol-3-O-rutinoside showed a good linear in the range of 5.11~51.07μg/mL(r=0.9999)and the average recovery was 98.0%,RSD was 1.1%(n=9).Conclusion The method was accurate,simple,and sensitive,which could be used for the quality control of the Nymphaea Candida.
