基于生物信息学分析TOP2A在肝癌中的表达和意义

摘要 目的:基于生物信息学分析肝细胞癌(liver hepatocelluar carcinoma,LIHC)组织中拓扑异构酶IIA(topoisomerase IIA,TOP2A)的表达和意义,探讨TOP2A在肝癌发生发展中的可能机制。方法:利用TCGA数据库肝癌组织中TOP2A的信息和GEPIA、UALCAN、cBioPortal等分析工具分析TOP2A基因表达与免疫浸润、预后的相关性。结果:肝癌组织中TOP2A基因的表达要高于正常组织,差异具有统计学意义(P<0.05);肝癌组织中TOP2A基因为低甲基化水平;TOP2A基因的表达与免疫细胞浸润正相关,且TOP2A基因高表达患者的总生存时间(overall survival,OS)、无病生存时间(disease-free survival,DFS)和生存率均低于TOP2A基因低表达患者(P<0.05)。结论:TOP2A基因可以作为肝癌预后的预测指标,TOP2A基因甲基化和免疫细胞的浸润可能在肝癌的发病和预后过程中具有重要意义。 Objective Based on bioinformatics analysis,the expression and significance of topoisomeraseⅡA(TOP2A)in liver hepatocelluar carcinoma(LIHC)were analyzed to explore the possible mechanism of TOP2A in the occurrence and development of liver cancer.Methods The correlation of TOP2A gene expression with immune infiltration and prognosis was analyzed by using the information of TOP2A in liver cancer tissue from the TCGA database and analysis tools such as GEPIA,UALCAN and cBioPortal.Results The expressions of TOP2A gene and protein in tumor tissues were higher than those in normal tissues,and the differences were statistically significant(P<0.05).TOP2A gene in liver cancer tissue was at low methylation level and TOP2A gene expression was positively correlated with immune cell infiltration.The overall survival(OS),disease-free survival(DFS)and survival rate of patients with high TOP2A gene expression were lower than those of patients with low TOP2A gene expression(P<0.05).Conclusion TOP2A gene can be used as a predictor for liver cancer prognosis.TOP2A gene methylation and immune cell infiltration may have important significance in the pathogenesis and prognosis of liver cancer.
