摘要
目的基于HPLC法测定车前子生品、炒制品、盐制品中京尼平苷酸和毛蕊花糖苷的含量,以比较它们之间的差异,为优选车前子优质饮片提供科学依据。方法色谱柱:RD-C18(250 mm×4.6 mm,5μm);以乙腈为流动相A,以0.1%磷酸水溶液为流动相B,梯度洗脱(0~7 min,14%A;7~27 min,14%~24%A);流速:1 mL/min;检测波长为254 nm;柱温:35℃;进样体积:10μL。结果京尼平苷酸在1.6~8μg范围内线性关系良好,毛蕊花糖苷在1.0~5.0μg范围内线性关系良好;生车前子经炮制后京尼平苷酸含量有所增加,而毛蕊花糖苷有所降低。结论该方法分离效果好,操作简单,方便易行,测定结果准确度高,可为车前子及其炮制品的质量评价提供依据。
Objective To determine the contents of Geniposide and verbascoside in Plantaginis Semen by HPLC and to compare the differences between them so as to provide scientific basis for the optimization of high quality pieces of Plantaginis Semen.Methods Chromatographic column is RD-C18(250 mm×4.6 mm,5μm).Acetonitrile was used as mobile phase A,and 0.1%phosphoric acid solution as mobile phase B.The mobile phase was degassed in an ultrasonic cleaning instrument and eluted in gradient(0~7 min,14%A,7~27 min,14%~24%A).Flow rate is 1 mL/min.The detection wavelength is 254 nm.The column temperature is 35℃.Injection volume is 10μL.Results The linear range of Geniposide is 1.6~8μg,while the verbascoside in 1.0~5.0μg.After processing,the content of Geniposide increased and the content of verbascoside decreased.Conclusion The method has good separation effect,simple operation,convenience and high accuracy,which could provide experimental basis for the quality evaluation of Plantaginis Semen and its processed products.
