大白菜新品种陕秋白3号种子纯度的分子鉴定 被引量:2

摘要 为了加快大白菜新品种陕秋白3号的推广,用分子标记技术进行种子纯度鉴定研究。从28对SSR引物中筛选出陕秋白3号的特异扩增引物SSRA 07-1和SSRA 07-2,这两对引物在两个亲本中扩增出不同带型,在杂交种中扩增出共显性产物,可以将杂交种和亲本有效区分开。鉴定结果为:正交杂交率96.7%,反交杂交率88.3%,与田间鉴定的正交杂交率96%,反交杂交率89%基本一致。进一步用这两对引物对陕秋白3号与秋白85、秦白二号、14杂1、27-2、金冠1号、改良青杂3号、秋早55等7个大白菜品种进行鉴定,结果显示,该引物可以将陕秋白3号与这7个品种有效区分开。研究表明,利用SSR鉴定可以快速鉴定新品种陕秋白3号,加快其在生产上的推广速度。 In order to speed up the popularization of a new Chinese cabbage variety‘Shaanqiubai 3’,we carried out a study on its seed purity identification using molecular marker technology.The specific amplification primers SSRA 07-1 and SSRA 07-2 of‘Shaanqiubai 3’were screened from 28 pairs of SSR primers,of which the two pairs of primers amplified different band types in the two parents,and amplified codominant products in the hybrid,which could effectively distinguish the hybrid from the parent.The identification results showed that the orthogonal hybridization rate was 96.7%,and the reverse hybridization rate was 88.3%,which was basically consistent with the field identification of the orthogonal hybridization rate of 96%and the reverse hybridization rate of 89%.Further using these two pairs of primers‘Shaanqiubai 3’and known‘Qiubai 85’,‘Qinbai 2’,‘14 Za 1’,‘27-2’,‘Jinguan 1’,‘Gailiang qingza 3’,and‘Qiuzao 55’,to be identified,and the results showed that the primers could effectively distinguish‘Shaanqiubai 3’from these 7 cultivars.Therefore,we concluded that the use of SSR identification could quickly identify the new variety‘Shaanqiubai 3’and speed up the promotion in production.
