糖化血红蛋白A1亚基在肺癌发生过程中的作用

摘要 目的:探讨糖化血红蛋白A1亚基(HbA1c)在肺癌的肿瘤发生中的作用及机制。方法:构建BALB/c裸鼠肺癌原位癌动物模型,采用皮下注射A549细胞的方式进行肺癌原位癌造模。采用实时荧光定量PCR(qRTPCR)技术检测细胞中血红蛋白A1亚基(HbA1)的表达,采用蛋白印迹法检测HbA1和HbA1c的表达。采用CCK-8实验检测A549细胞的增殖速度,采用Transwell小室实验检测A549细胞的迁移能力。结果:肺癌原位癌小鼠血清中糖化血红蛋白的表达水平显著高表达。糖化血红蛋白HbA1c上调后A549细胞的增殖和迁移能力显著上调,呈现显著的癌化趋势。小鼠HbA1c上调后小鼠肺癌原位癌肿瘤显著增大,促进肿瘤的发生。结论:初步发现糖化血红蛋白A1亚基能促进肺癌的增殖和迁移。 Objective:To investigate the effect and mechanism of HbA1c in the tumorigenesis of lung cancer in mice.Methods:An animal model of lung cancer in situ carcinoma in BALB/c mice was constructed by subcutaneous injection of A549 cells.The expression of HbA1c was detected by qRT-PCR.The expression of hemoglobin HbA1 and HbA1c was detected by Western Blot.The proliferation rate of A549 cells was measured using the CCK-8 assay,and the cell migration ability of A549 cells was measured using the transwell assay.Results:The expression level of glycosylated hemoglobin in the serum of mice with lung cancer in situ was significantly raised.The proliferation and migration ability of A549 cells was significantly up-regulated after HbA1c up-regulation,presenting a significant carcinogenic trend.After the up-regulation of HbA1c in mice,the lung cancer tumors in mice were significantly enlarged,and the occurrence of tumors was promoted.Conclusion:Our research provided a preliminary view that HbA1c could promote the proliferation and migration of lung cancer.
