以人脐带间充质干细胞为种子细胞促进脂肪移植存活率的研究

摘要 目的本实验将具有多向分化潜能的人脐带间充质干细胞(hUC-MSCs)作为种子细胞,诱导分化为组织工程脂肪,探讨其在脂肪移植中提高脂肪存活率的能力。方法选取健康雄性BALB/c裸鼠30只,随机分为7、14、28 d组,每组10只;选取健康雄性SD大鼠10只,脱椎处死后按照常规方法提取肠系膜、腹股沟处脂肪颗粒。培养hUC-MSCs并进行成脂诱导后混合大鼠脂肪颗粒移植于3组裸鼠左侧腹部筋膜下作为实验侧,将相同剂量的单纯脂肪颗粒移植于右侧腹部筋膜下作为对照侧。移植后对3组裸鼠的移植脂肪进行组织病理学检测,并比较移植脂肪组织的质量维持率。结果7、14及28 d组裸鼠间,实验侧移植脂肪组织质量维持率逐渐降低(F=58.47,P<0.05),对照侧移植脂肪组织质量维持率也逐渐降低(F=225.28,P<0.05);在3组裸鼠中,实验侧移植脂肪组织质量维持率均显著高于对照侧(F=1490.66~2142.95,P<0.05)。结论hUC-MSCs来源的脂肪细胞与天然的脂肪细胞颗粒混合后移植可以有效提高移植后脂肪细胞的存活率。本研究为以hUC-MSCs作为种子细胞的组织工程脂肪在脂肪移植中的应用奠定了实验基础。 Objective To explore the ability to improve fat graft survival of multipotent human umbilical cord mesenchymal stem cells(hUC-MSCs)as seed cells to derive adipose tissue in fat transplantation.Methods Thirty healthy male BALB/c nude mice were randomly divided into 7 d,14 d,and 28 d groups,with 10 mice in each group.Ten healthy male SD rats were sacrificed by cervical dislocation to obtain fat granules in the mesentery and inguinal region using conventional methods.The fat granules were mixed with hUC-MSC cultures that had been treated with adipogenic induction,and were then transplanted into each nude mouse’s left subfascial abdominal area(experimental side),with the same dose of fat granules alone transplanted into the right subfascial abdominal area(control side).Histopathological examination was performed to compare the weight maintenance of fat grafts between the three groups.Results The weight maintenance rate of fat grafts was significantly decreased with the time of grafting(the 7 d group>the 14 d group>the 28 d group)for both the experimental side(F=58.47,P<0.05)and the control side(F=225.28,P<0.05).For all the three groups,the weight maintenance rate of fat grafts on the experimental side was significantly higher than that on the control side(F=1490.66-2142.95,P<0.05).Conclusion hUC-MSCs-derived adipocytes mixed with natural adipocyte granules can effectively improve the survival of adipocytes after transplantation,laying an experimental foundation for the application of hUC-MSCs-derived adipocytes in fat transplantation.
