基于高通量培养组学的人体皮肤细菌多样性研究 被引量:1

Bacterial diversity of human skin based on high-throughput culturomics

摘要 了解中国健康人皮肤可培养细菌多样性及建立中国健康人皮肤细菌菌株资源库,能够为皮肤细菌研究及研究皮肤细菌与皮肤疾病之间的潜在关系提供菌株资源。通过高通量培养组学技术,利用9种培养基对18名健康中国人,8个部位,144份皮肤样品进行高通量分离培养,并对筛选获得的细菌进行16S rRNA测序鉴定分析。研究表明,共分离获得3580株细菌,隶属于4门33属108种;4个门包括厚壁菌门(Firmicutes)、放线菌门(Actinobacteria)、变形菌门(Proteobacteria)和拟杆菌门(Bacteroidetes),其中优势门为厚壁菌门(占总菌株数的87.01%),其次为放线菌门(10.61%);优势属为葡萄球菌属(Staphylococcus)(占总菌株数的82.07%);优势种为表皮葡萄球菌(Staphylococcus epidermidis)(占总菌株数的35.81%),其次为人葡萄球菌抗新霉素败血症亚种(Staphylococcus hominis subsp.novobiosepticus)(26.37%)及头状葡萄球菌(Staphylococcus capitis)(9.44%);并获得39株26种潜在的新菌种,其与最接近的16S rRNA序列相似率分布为88.26%~98.61%,潜在新属新种占总菌株的1.09%,占所有菌种的24.07%。健康中国人群皮肤细菌资源丰富,且存在大量的潜在新属新种有待分离培养鉴定;我们初步建立了健康中国人皮肤细菌资源库,以丰富现有皮肤细菌资源,为利用皮肤细菌资源提供菌株保障。 This study is to explore the diversity of culturable bacteria in the skin of Chinese healthy people and establish a resource bank of Chinese healthy human skin bacterial strains,so as to provide strain resources for the study of skin bacteria and the protential relationship between skin bacteria and skin diseases.We collected 144 skin samples from 8 different skin sites of 18 healthy Chinese,and performed high-throughput isolation and culturation of bacteria from these samples using 9 different culture media.The isolated bacteria were identified according to their 16S rRNA sequences.A total of 3580 bacterial strains were isolated in this study.These isolates are assigned to 108 species of 33 genera and 4 phyla.Among the four phyla(Firmicutes,Actinobacteria,Proteobacteria and Bacteroidetes),Firmicutes(87.01%)is the dominant phylum,followed by Actinobacteria(10.61%).The dominant genus is Staphylococcus(82.07%).Staphylococcus epidermidis(35.81%)is the dominant species,followed by Staphylococcus hominis subsp.novobiosepticus(26.37%)and Staphylococcus capitis(9.44%).Meanwhile,26 potential novel species were identified for 39 bacterial strains,among which the similarities of 16S rRNA sequences to the most closest sequences deposited in the public database were 88.26%~98.61%.The potential novel genera and species accounted for 1.09%of all strains,and 24.07%of all species.Chinese healthy people are rich in skin bacterial resources,and there are a large number of potential new genera and species to be isolated and identified.We have initially established a healthy human skin bacterial resource bank to enrich the existing skin bacterial resources and provide strain guarantee for the use of skin bacterial resources.
