摘要 目的:建立栝楼离体组织培养高效再生体系。方法:以栝楼茎尖为材料,MS为基本培养基,研究不同消毒时间对茎尖再生苗的影响,以及不同激素水平对茎尖再生芽数及茎段增殖、生根的影响。结果:最适茎尖外植体消毒组合为:75%酒精40 s+0.1%升汞6 min,污染率及死亡率低,再生苗成苗率高;茎尖再生芽数的影响因素主次顺序为6-BA>IBA>NAA>KT;最适茎段增殖激素配方为0.4 mg/L 6-BA和0.06 mg/L NAA,增殖倍数、株高分别为5.42、2.12 cm,茎节正常伸长、叶色淡绿;最适生根激素浓度为1.0 mg/L IBA,生根率为92.2%,根数为5.3个,根长为7.6 cm,叶淡绿,苗较壮。结论:通过相关激素浓度筛选试验,建立了栝楼离体组织培养再生体系,并获得了大量无菌再生种苗。 Objective:To establish the regenerative system of Trichosanthes kirilowii Maxim in vitro.Methods:The shoot tips of Trichosanthes kirilowii Maxim were chosen as explants for plant tissue in the MS medium.The effects of different disinfection time on shoot tip regenerated seedlings were studied at first.And then,the effects of different concentrations of hormones on stem apex regenerated buds,shoot proliferation and rooting were investigated.Results:75% alcohol treated for 40 s and 0.1% HgCl;treated for 6 min were suitable for explants sterilization.The combination got lower pollution and mortality,and higher plantlet regeneration rates.The order of influencing factors on shoot tips regeneration number was 6-BA>IBA>NAA>KT.The optimal hormone formula of shoot proliferation were 0.4 mg/L 6-BA and 0.06 mg/L NAA,and the coefficient of proliferation was 5.42,the plant height was 2.12 cm,stem nodes length was normal and the leaves were green.The optimal hormone concentration of rooting was 0.01 mg/L IBA,and the rooting rate was 92.2%,the number of roots was 5.3,the rooting length was 7.6 cm,the leaves were green,and the seedlings were strong.Conclusion:The regeneration system of Trichosanthes kirilowii Maxim in vitro was established by screening the relevant hormone concentration,and a large number of aseptic regeneration seedlings were obtained.
