摘要
目的通过研究患者导尿管样品中微生物群落构成,探索感染相关的潜在风险因素。方法收集2020年11~12月期间西苑医院38例患者留置的导尿管。提取样品中的细菌总DNA,PCR扩增16S rRNA V4-V5区。利用Hiseq 2500测序平台进行测序,数据使用skewer、FLASH等软件进行细菌群落结构及多样性分析。结果38例患者平均年龄为(82.1±7.2)岁,导尿管平均留置时间为(25.50±9.66)d。有效序列1603007条,有效序列长度在382bp至442bp之间,共得到11350个OTUs和1,668,076个优质序列。S1-S5时间段分组,各组样品中革兰氏阴性菌所占比例均高于阳性菌,36个样品存在含量>50%的优势菌门,占全部样品的86.8%。结论导尿管为相对封闭环境,微生物群落具有特定性,导尿管中菌群变化规律总体上与留置时间相关,长留置时间可能导致高度单一菌群结构出现。
Objective This study focused on the composition of microbial community of patients’catheter sample to explore the potential risk factors of urinary tract infection.Methods A total of 38 catheter samples were collected from patients in Xiyuan Hospital during November to December 2020 in Beijing.Total bacterial DNA was extracted from specimens,and V4-V5 region of 16S rRNA was amplified by PCR.Hiseq 2500 sequencing platform was used for sequencing,and skewer and FLASH were used to analysis the structure and diversity of bacterial community.Results The average age of 38 patients was(82.1±7.2)years,and the average indwelling time of catheter was(25.50±9.66)d.There were 1603007 effective sequences,the length of effective sequences ranged from 382bp to 442bp,a total of 11350 OUTs and 1668076 high quality sequences were obtained.The proportion of gram-negative bacteria in all S1-S5 groups was higher than gram-positive bacteria.Thirty six samples had dominant phylum with content>50%,accounting for 86.8%.Conclusion Catheter is a relatively closed environment,the microbial community is specific in this situation,and the change of microbial community is related to the retention time.Long retention time may lead to the appearance of highly single bacteria structure.
