摘要
目的观察交泰丸对心肾不交失眠模型大鼠睡眠质量的影响。方法21只SD大鼠随机分为空白组、模型组、交泰丸组,每组7只。除空白组外,其余各组大鼠采用复合刺激+腹腔注射对氯苯丙氨酸(PCPA)建立心肾不交失眠大鼠模型,造模后交泰丸组给予交泰丸1.1 g/(kg·d)灌胃,空白组和模型组给予蒸馏水10 ml/(kg·d)灌胃,各组均灌胃7天。通过动物睡眠生物解析系统记录脑电(EEG)和肌电(EMG)信号24 h,分析大鼠夜晚(19:00~次日7:00)和白天(次日7:00~19:00)觉醒(Wake)量、非快速眼动(NREM)睡眠总量和快速眼动(REM)睡眠总量,各睡眠时相的转换次数,不同时程NREM睡眠、REM睡眠和Wake出现次数,脑电频谱密度;记录结束后测定大鼠肾上腺指数,ELISA法检测大鼠血清促肾上腺皮质激素(ACTH)、促肾上腺皮质激素释放激素(CRH)含量。结果与空白组比较,模型组大鼠肾上腺指数升高、血清ACTH含量升高(P<0.05或P<0.01),但CRH含量差异无统计学意义(P>0.05);夜晚和白天NREM睡眠总量均减少,REM睡眠总量减少,Wake总量增多(P<0.05或P<0.01),主要表现为10~20 s和30~50 s NREM睡眠次数显著增多,120~230 s和240~470 s NREM睡眠次数显著减少(P<0.05或P<0.01),NREM睡眠转换为Wake的次数增加,NREM睡眠转换为REM睡眠的次数减少,REM睡眠转换为Wake的次数减少(P<0.05或P<0.01)。与模型组比较,交泰丸组肾上腺指数下降、血清ACTH含量下降(P<0.05),但CRH差异无统计学意义(P>0.05);夜晚和白天NREM睡眠总量均显著增加,Wake总量均显著减少(P<0.01),主要表现为10~20 s和30~50 s NREM睡眠次数显著减少,120~230 s和240~470 s NREM睡眠次数显著增加(P<0.01),NREM睡眠与Wake之间的转换次数减少(P<0.01)。与空白组比较,交泰丸组大鼠NREM睡眠的Delta波密度和REM睡眠的Theta波密度差异无统计学意义(P>0.05)。结论交泰丸主要通过增加较长时程(120~230 s、240~470 s)NREM睡眠,减少NREM睡眠与Wake之间的转换次数,提高睡眠时间和睡眠质量,且诱导的睡眠类似于生理性睡眠。
ObjectiveTo evaluate the effect of Jiaotai Pills(交泰丸)on the sleep stage and sleep quality in insomnia model rats with syndrome of non-interaction between the heart and the kidney.Methods rats were randomly divided into control group,model group and Jiaotai Pills group,with 7 rats in each group.Except for the control group,the rat model of insomnia with non-interaction between the heart and the kidney was established through compound stimulation and intraperitoneal injection of para-chlorophenylalanine(PCPA).After modeling,the Jiaotai Pills group was administrated with 1.1 g/kg of Jiaotai Pills by gavage,while the control group and model group were given 10 mL/kg of distilled water by gavage,once a day for 7 consecutive days.Electroencephalogram(EEG)and electromyogram(EMG)were recorded through animal sleep bioanalytical system.The total amount of night[(19:00-07:00(+1)]and day[(07:00(+1)-19:00(+1)]wake,non-rapid eye movement(NREM)sleep and rapid eye movement(REM)sleep,the number of sleep stage transitions,the episode number of NREM sleep,REM sleep and wake,and EEG power density were analyzed.After recording,the adrenal index was determined,and the adrenocorticotropic hormone(ACTH)and corticotropin-releasing hormone(CRH)levels in serum were detected through ELSA.Results model group significantly increased(P<0.05 or P<0.01),while there was no difference between groups in CRH level(P>0.05);the total amount of night and day NREM sleep and REM sleep were reduced,while the wake amount increased(P<0.05 or P<0.01),with increased episode number of 10-20 s and 30-50 s NREM sleep and decreased 120-230 s and 240-470 s NREM sleep(P<0.05 or P<0.01),as well as increased number of NREM sleep transiting into wake and decreased number of NREM sleep transiting into REM sleep and that of REM sleep transiting into wake(P<0.05 or P<0.01).Compared to those in the model group,the adrenal index and ACTH level in serum in the Jiaotai Pills group were reduced(P<0.05),but there was no difference in CRH(P>0.05);the total amount of night and day NREM sleep increased while the wake amount was reduced significantly(P<0.01),with decreased episode number of 10-20 s and 30-50 s NREM sleep and increased episode number of 120-230 s and 240-470 s NREM sleep(P<0.01),as well as reduced number of NREM sleep transiting into wake(P<0.01).There were no significant differences in the Delta wave density of NREM sleep and the Theta wave density of REM sleep between the Jiaotai Pills group and control group(P>0.05).Conclusion Jiaotai Pillscan improve sleep stage and sleep quality mainly by increasing long duration NREM sleep(120-230 s and 240-470 s)and reducing the transition number between NREM sleep and wake,and the induced sleep is similar to physiological sleep.
