摘要 目的 确定细胞焦亡相关基因与肾透明细胞癌患者的预后之间的关联。方法 发现有24个与细胞焦亡有关的基因在肾透明细胞癌和正常肾脏组织之间有不同的表达。使用这些差异表达基因通过共识聚类分析将肾透明细胞癌病例分成两个亚组,然后使用LASSO Cox回归模型建立基于癌症基因组图谱队列的多基因预后模型。因此,建立了一个对肾透明细胞癌患者具有良好预测能力的由11个基因组成的模型(P<0.000 1)。结果 KEGG和GO分析结果显示了免疫相关基因的富集,高风险组的病人相对于低风险组患者,高风险组患者的免疫状态明显降低。结论 这些发现表明与细胞焦亡相关的基因是预测肾透明细胞癌预后的重要因素,并且这些基因在肿瘤免疫中发挥关键作用。 Objective The objective of this article is to determine an association between the pyroptosis-associated genes and the prognosis of patients with KIRC. Methods We used these DEGs to stratify KIRC cases into two subgroups, through consensus clustering, then build a multigene prognostic signature based on a The Cancer Genome Atlas(TCGA)cohort using the least absolute shrinkage and selection operator(LASSO) cox regression model. Consequently, we established an 11-gene signature that exhibited favorable predictive power of KIRC patients(P<0.000 1). Results Results from the Kyoto Encyclopedia of Genes and Genomes(KEGG) and gene ontology(GO) analyses showed enrichment of immune-associated genes, with high-risk group patients exhibiting significantly low immune status relative to low-risk group patients. Conclusions These findings indicate that pyroptosis-associated genes are important predictors of prognosis in KIRC and play crucial roles in tumor immunity.
