掺钒雪菊的制备及其对α-葡萄糖苷酶活性抑制研究

摘要 目的:以掺杂的方法制备掺钒雪菊,探究雪菊富钒的可行性,并考察其提取物对α-葡萄糖苷酶活性的影响及其酶抑制作用类型。方法:采用石墨炉原子吸收分光光度法测定钒(V)含量,并计算雪菊富钒率;以4-硝基苯基-α-D-吡喃葡萄糖苷(pNPG)为底物,阿卡波糖为阳性对照,测定提取物对α-葡萄糖苷酶活性的抑制率并进行抑制动力学分析。结果:经测定,雪菊富钒能力较强,钒化合物浓度在25~800mg/L范围内,雪菊富集钒总量在1.44~30.82mg/kg之间。当钒溶液浓度为50mg/L时,富钒率最高;掺钒雪菊提取物对α-葡萄糖苷酶的半数抑制浓度(IC_(50))为67.08±1.15μg/mL,且抑制率随浓度增加而增加。酶抑制动力学实验研究发现其提取物为混合型抑制类型。结论:在适宜的钒浓度条件下,掺钒雪菊提取物具有较高的富钒率和较强的酶抑制活性。 Objective:Vanadium-doped in Coreopsis tinctoria Nutt.was prepared by artificial doping Vanadium method.The Vanadium enrichment capacity in C.tinctoria and its extraction to inhibit α-glucosidase activity and the type of enzyme inhibition was also investigated.Methods:The content of Vanadium was determined by graphite furnace atomic absorption spectrophotometer and the vanadium enrichment rate was calculated in C.tinctoria.The α-glucosidase inhibitory activity was determined.Using 4-nitrophenyl-α-D-glucopyranoside(PNPG)as substrate and acarbose as positive control and the inhibition kinetics was analyzed.Results:The results showed that the Vanadium was enriched in C.tinctoria by artificial doping Vanadium method.The total amount of Vanadium in C.tinctoria,was at 1.44~30.82 mg/kg when added vanadium in the range of 25~800 mg/L concentration.The ratio of enrichment vanadium was the highest when the concentration of vanadium solution at 50 mg/L.The α-glucosidase inhibitory activity IC_(50) was 67.08±1.15μg/m L,and theα-glucosidase inhibitory capacity of the enrichment Vanadium extraction increased in a dose-dependent manner.The enzyme inhibition kinetics experiment showed that the extracts were mixed inhibition type.Conclusion:The high Vanadium enrichment capacity and α-glucosidase inhibitory activity of C.tinctoria extraction will be obtained at the suitable concentration of artificial doping Vanadium solution.
