高效液相色谱法测定黄原胶乙酰基含量 被引量:1

摘要 黄原胶分子结构上乙酰基化在一定程度上影响了产品性能,为更好地研究黄原胶性能与应用,通过实验验证建立了一种高效液相色谱法测定黄原胶乙酰基含量的方法。黄原胶样品脱乙酰基处理最优条件为60℃处理3h;检测色谱条件为:安捷伦HI-Plex H色谱柱,高效液相色谱UV检测器,检测波长为210nm;以5mmol/L硫酸水溶液为流动相,流速为0.6mL/min,柱温60℃,进样体积20μL。方法验证实验结果表明:方程线性关系良好,相关系数0.99998,定量检出限为0.165mg/(100mL)。测得乙酰基含量数据RSD值为1.1%,样品处理液在48h内稳定,加标回收率为97.4%。 The acetylation of the molecular structure of xanthan gum affects the performance of the product to a certain extent.In order to better study the performance and application of xanthan gum,a high performance liquid chromatography method was established to determine the content of xanthan gum.The optimal conditions for deacetylation treatment of xanthan gum samples was 60℃for 3 hours.The detection chromatographic conditions were:Agilent chromatographic column(HI-Plex H)with 5mmol/L sulfuric acid aqueous solution as the flow Phase at a flow rate of 0.6mL/min,the detection wavelength of UV detector was 210nm;the column temperature was 60℃,and the injection volume was 20μL.The results of the validation tests for the method showed that the correlation coefficient of the equation with well linearity was 0.99998,and the quantitative detection limit was 0.165mg/(100mL).The RSD value of the content data was 1.1%,the sample treatment solution was stable within 48 hours,and the recovery rate of standard addition was 97.4%.
