利多卡因对七氟醚致发育期大鼠认知功能的影响

摘要 目的 通过观察利多卡因对七氟醚致发育期大鼠认知功能的影响,进一步研究利多卡因在调节七氟醚致发育期大鼠海马组织自噬中的作用。方法 清洁级健康SD大鼠36只,雌雄不限,7日龄,按随机分组原则分为三组(n=12)。七氟醚+利多卡因组:(S+L组)出生第7-9 d腹腔注射利多卡因10 mg/kg后行七氟醚暴露;对照组:(C组)出生第7-9 d腹腔注射同体积生理盐水后行混合气体暴露;七氟醚组:(S组)出生第7-9 d腹腔注射同体积生理盐水后行七氟醚暴露。待大鼠长至20 d时行水迷宫实验,实验后麻醉处死大鼠,取海马组织。免疫印迹法检测海马组织中自噬蛋白LC3、BECN1、P62相对表达量。结果 与C组相比,S组和S+L组逃避潜伏期延长,平台象限停留时间缩短,LC3,BECN1相对表达量增加,P62表达减少;与S组相比,S+L组逃避潜伏期缩短,平台象限停留时间延长,LC3,BECN1相对表达量减少,P62表达增加。结论 利多卡因对七氟醚致发育期大鼠认知功能障碍有保护作用,其机制可能与减少过度自噬有关。 Objective To observe the effect of lidocaine on the cognitive function of sevoflurane-induced developmental rats, and to further study about the role of lidocaine in regulating sevoflurane-induced mitochondrial autophagy in the hippocampus of developmental rats. Methods Thirty-six Sprague Dawley healthy rats(male or female, 7 days old)were randomly divided into the Control group(group C), Sevoflurane group(group S)and Sevoflurane combined with lidocaine group(Group S+ L), with 12 rats in each group. In group S+ L,10 mg/kg lidocaine was intraperitoneally injected, and sevoflurane were given at 7-9 days after birth. Group C were all dealt with intraperitoneal injection of the same volume of normal saline and then exposed to a mixture of air and oxygen at 7-9 days after birth. Group S were dealt with intraperitoneal injection of the same volume of normal saline, and then exposed to sevoflurane at 7-9 days after birth.Morris water maze training were carried out when the rats grew to 20 days after birth. After that, the rats were killed after anesthesia and the hippocampus tissue was taken. Western-blot experiment was used to detect the relative expression of autophagy proteins LC3, BECN1,P62 in the hippocampus. Results Compared with Group C, the escape latency of group S and group S+L was prolonged, the residence time in the quadrant of platform was shortened, the relative expression of LC3, BECN1 increased, the expression of P62 decreased.Compared with the Group S, the Group S+L had a shorter escape latency and a longer stay in the quadrant of platform, the relative expression of LC3, BECN1 decreased, the expression of P62 increased. Conclusions Lidocaine has a positive effect on sevoflurane-induced cognitive dysfunction in rats, and its mechanism may be related to the reduction of excessive autophagy.
