摘要 目的观察维生素D对脱氢表雄酮(DHEA)和高脂饮食诱导多囊卵巢综合征(PCOS)小鼠模型的作用。方法将40只青春前期雌性小鼠随机分为4组:对照组,PCOS组,低剂量Vit.D组,高剂量Vit.D组。对照组小鼠普通饲料喂养,皮下注射溶剂,其余组小鼠采用每天皮下注射6 mg/kg DHEA并给予高脂饮食至试验终止的方法建立PCOS模型。第31天开始,对照组和PCOS组每周腹腔注射大豆油,低剂量Vit.D组腹腔注射1,25(OH)2D31.3μg/(kg·周),高剂量Vit.D组腹腔注射1,25(OH)2D313μg/(kg·周)。第70天,采集各组小鼠血清及肝脏组织。结果高剂量Vit.D组小鼠在注射维生素D后2周内全部死亡。PCOS组小鼠体重及睾酮水平显著高于对照组和低剂量Vit.D组(P<0.05)。对照组、PCOS组和低剂量Vit.D组小鼠肝重/体重值有显著差异(P<0.05)。结论维生素D可以改善PCOS的高雄激素血症及肥胖,可能通过调节肝的代谢功能来实现,而过高剂量的维生素D摄入可能有害。 Objective This study aimed to investigate effects of vitamin D on dehydroepiandrosterone(DHEA)and high fat diet PCOS mouse models.Methods Prepubertal female mice were randomly divided into 4 groups:the control group,PCOS group,low dose Vit.D group and high dose Vit.D group(n=10 per group).Controls were injected with vehicle alone and fed with normal diet.The mice in the other three groups were subcutaneously injected with 6mg/kg/day DHEA and fed with high fat diet.After 30 days,Vit.D was administrated by intraperitoneal injection in the following 40 days,1.3μg/kg/week 1,25(OH)2D3 in low dose Vit.D group,and 13μg/kg/week 1,25(OH)2D3 in high dose Vit.D group.Controls also accepted intraperitoneal injections but with vehicle alone.At the end of the 70 days,blood samples were collected and the liver tissues were taken.Results All the mice in high dose Vit.D group died in two weeks after Vit.D administration.The body weight and testosterone levels in PCOS group were significantly higher than those in control group and low dose Vit.D group(P<0.05).The ratio of liver to body weight was significantly different among the three groups(P<0.05).Conclusion Our results indicate that Vit.D has positive effects on obesity and hyperandrogenemia,maybe through liver metabolism regulation.High dose Vit.D administration may be harmful.
