烟曲霉菌感染后炎症与抗炎因子动态变化及骨髓间充质干细胞移植的干预作用

Dynamic changes of inflammation and anti-inflammatory factors after Aspergillus fumigatus infection and intervention of bone marrow mesenchymal stem cell transplantation

摘要 目的观察烟曲霉菌感染大鼠血浆炎症与抗炎因子动态变化,及骨髓间充质干细胞移植(BMD-MSCs)干预的保护作用。方法72只雄性SD大鼠随机分为对照组、曲霉菌感染组、BMD-MSCs干预组;曲霉菌感染成功后即刻经尾静脉注10E6(BMD-MSCs),分时处死实验动物,检测血浆细胞因子及肺组织HE染色。结果曲霉菌感染组较对照组的α肿瘤坏死因子(TNF-α)、白介素-6(IL-6)、白介素-10(IL-10)、烟曲霉特异性IgG、总IgE浓度明显升高,差异有统计学意义(P<0.05),BMD-MSCs干预组较曲霉菌感染组的TNF-α、IL-6、烟曲霉特异性IgG、总IgE浓度浓度低,差异有统计学意义(P<0.05);BMD-MSCs干预组较曲霉菌感染组的IL-10浓度高,差异有统计学意义(P<0.05)。烟曲霉菌感染后肺间质水肿、肺泡结构破坏、48 h组更明显;BMD-MSCs干预组较曲霉菌感染组有不同程度改善,间质水肿减轻。结论BMD-MSCs对烟曲霉菌感染大鼠具有保护作用;BMD-MSC可以升高抗炎因子IL-10及降低促炎因子TNF-α、IL-6水平。 Objective To observe the dynamic changes of plasma inflammatory factors and anti-inflammatory factors in rats withAspergillus fumigatusinfection and the protective effect of bone marrow mesenchymal stem cell transplantation(BMD-MSCs).Methods Seventy-two male SD rats were randomly divided into blank control group(group K),Aspergillus fumigatusinfection group(group G),BMD-MSCs intervention group(group GH).10E6(BMD-MSCs)was injected via tail vein immediately,and the animals were killed at different times.The plasma was taken for cytokine detection and lung tissue HE staining.Results The concentrations of TNF-α,IL-6,IL-10,Aspergillus fumigatusspecific IgG and total IgE inAspergillus fumigatusinfection group were significantly higher than those in blank control group,with the differences statistically significant(P<0.05).The concentrations of TNF-α,IL-6,Aspergillus fumigatusspecific IgG and total IgE in group GH were lower than those in group G,with the differences statistically significant(P<0.05).The concentrations of IL-10 in BMD-MSCs intervention group were higher than those inAspergillus fumigatusinfection group,with the difference statistically significant(P<0.05).AfterAspergillus fumigatusinfection,pulmonary interstitial edema and alveolar structure destruction were more obvious in 48 h.Compared withAspergillus fumigatusinfection group,BMD-MSCs intervention group had different degrees of improvement,and interstitial edema was reduced.Conclusion BMD-MSCs have protective effect on Aspergillus infection in rats,which can increase the level of IL-10 and decrease the levels of TNF-αand IL-6.
