摘要
目的 探讨miR-671-3p靶向调控细胞骨架相关蛋白4(CKAP4)对人脑胶质瘤细胞增殖及迁移的影响。方法 收集术后脑胶质瘤组织及配对癌旁正常组织标本,检测其miR-671-3p和CKAP4表达水平。将人脑胶质瘤U251细胞株分为miR-671-3p NC组、miR-671-3p mimic组和miR-671-3p inhibitor组,并转染相应质粒。采用实时荧光定量PCR测定miR-671-3p表达,CCK-8实验测定细胞增殖能力,Transwell实验测定细胞迁移能力。Western blotting测定CKAP4表达水平。结果 人脑胶质瘤组织中miR-671-3p、CKAP4表达水平高于癌旁正常组织(P<0.05)。细胞增殖率、迁移率miR-671-3p mimic组高于miR-671-3p NC组,miR-671-3p inhibitor组低于miR-671-3p NC组(P<0.05)。miR-671-3p对CKAP4具有靶向调节作用。miR-671-3p mimic组CKAP4蛋白表达量低于miR-671-3p NC组,miR-671-3p mimic组低于miR-671-3p inhibitor组(P<0.05)。结论 miR-671-3p可靶向调控CKAP4促进人脑胶质瘤细胞增殖及迁移能力。
Aim To explore the effect of miR-671-3 p targeting and regulating cytoskeleton-associated protein 4(CKAP4) on the proliferation and migration of human glioma cells. Methods Collect glioma tissues and paired normal tissue samples adjacent to cancer after surgery in our hospital to detect the expression levels of miR-671-3 p and CKAP4. Randomly divide the human glioma U251 cell line into miR-671-3 p NC group, miR-671-3 p mimic group and miR-671-3 p inhibitor group, the corresponding plasmids were transfected. Real-time fluorescent quantitative PCR experiment was used to measure the expression of miR-671-3 p, the CCK-8 experiment was used to measure the cell proliferation ability, and the Transwell experiment was used to measure the cell migration ability. Western blotting experiments to determine the expression level of CKAP4. Results The expression levels of miR-671-3 p and CKAP4 in human glioma tissues were higher than those in normal tissues adjacent to cancer(P<0.05). The cell proliferation rate and migration rate of miR-671-3 p mimic group were higher than miR-671-3 p NC group, and miR-671-3 p inhibitor group was lower than miR-671-3 p NC group(P<0.05). miR-671-3 p has a targeted regulatory effect on CKAP4. In the miR-671-3 p mimic group, the CKAP4 protein expression was lower than that of the miR-671-3 p NC group, and the miR-671-3 p mimic group was lower than the miR-671-3 p inhibitor group(P<0.05). Conclusion miR-671-3 p can target and regulate CKAP4 to promote the proliferation and migration of human glioma cells.
