ZBED3对血管内皮细胞增殖和迁移影响及其机制

The effect of ZBED3 on the proliferation and migration of vascular endothelial cells and its mechanism

摘要 目的探索ZBED3对血管内皮细胞HUVEC增殖和迁移能力的影响。方法转染ZBED3质粒和siRNA双向调控HUVEC中ZBED3的表达,采用MTT方法检测转染和干扰ZBED3对HUVEC增殖能力的影响;采用Transwell实验检测转染和干扰ZBED3对HUVEC迁移能力的影响;采用Western blot方法检测ZBED3在HUVEC中对经典Wnt信号通路活性的影响。结果转染ZBED3质粒能够显著增强HUVEC的增殖和迁移能力(P<0.05);siRNA干扰ZBED3能够显著抑制HUVEC的增殖和迁移能力(P<0.05);ZBED3能够上调HUVEC中β-catenin、cmyc和cyclin D1的表达,活化Wnt信号通路。结论ZBED3能够通过促进经典Wnt信号通路的活化,增强血管内皮细胞HUVEC的增殖和迁移能力。 Objective To explore the effect of ZBED3 on the proliferation and migration of vascular endothelial cells HUVEC.Methods Transfection of ZBED3 plasmid and siRNA respectively up-regulated and down-regulatedthe expression of ZBED3 in HUVEC.MTT method was used to detect the effect of ZBED3 transfection and interference on the proliferation of HUVEC.Transwell method was used to detect the effect of ZBED3 transfection and interference on the migration ability of HUVEC.Western blot method was used to detect the effect of ZBED3 on the activity of canonical Wnt signaling pathway in HUVEC.Results Transfection of ZBED3 plasmid significantly enhanced the proliferation and migration of HUVEC(all P<0.05);ZBED3 siRNA interference significantly inhibited the proliferation and migration of HUVEC(all P<0.05).ZBED3 can up-regulate the expression ofβ-catenin,c-myc and cyclin D1 in HUVEC and activate the Wnt signaling pathway.Conclusion ZBED3 could enhance the proliferation and migration of HUVEC of vascular endothelial cells by promoting the activation of the canonical Wnt signaling pathway.
