摘要
目的探讨微小RNA-181a-5p对白血病HL-60细胞增殖和侵袭的影响,并初步分析其机制。方法体外培养人白血病HL-60细胞,将miR-181a-5p抑制物转染白血病HL-60细胞,MTT法检测各组HL-60细胞的增殖能力,Transwell实验检测各组HL-60细胞的侵袭能力。实时定量PCR和Western blot检测各组细胞miR-181a-5p和PTEN的表达,双荧光素酶报告基因实验分析miR-181a-5p和PTEN的靶向关系。结果MTT和Transwell实验结果显示,转染miR-181a-5p抑制物后,HL-60细胞的增殖和侵袭能力下降(P<0.05),miR-181a-5p的表达下降,PTEN的表达上升(P<0.05)。双荧光素酶实验表明PTEN是miR-181a-5p的靶基因。结论抑制miR-181a-5p使白血病细胞HL-60的增殖和侵袭能力下降,其机制可能与上调PTEN表达有关。
Objective To investigate the effect and mechanism of miR-181 a-5 p on the proliferation and invasion of human leukemia HL-60 cells.Methods The human leukemia HL-60 cells were cultured,and the miR-181 a-5 p inhibitor was transfected into HL-60 cells.The proliferation ability of HL-60 cells was detected by MTT,and the invasion ability of HL-60 cells was detected by Transwell assay.The expression of miR-181 a-5 p and PTEN was detected by quantitative real-time PCR and Western blot.The target relationship between miR-181 a-5 p and PTEN was verified by double luciferase assay.Results MTT and Transwell results showed that after transfection of miR-181 a-5 p inhibitor,the proliferation and invasion ability of HL-60 cells was decreased(P<0.05),the expression of miR-181 a-5 p in HL-60 cells was decreased,and the expression of PTEN was increased.Dual luciferase assay showed that PTEN was the target gene of miR-181 a-5 p.Conclusion Inhibition of miR-181 a-5 p decreased the proliferation and invasion of HL-60 cells,which might be related to up-regulation of PTEN expression.
