仙灵骨葆胶囊对大鼠膝骨关节炎的治疗作用及机制 被引量:2

Therapeutical effect and mechanism of Xian-ling-gu-bao capsule on knee osteoarthritis in rats

摘要 目的探究仙灵骨葆胶囊对大鼠膝骨关节炎的治疗作用及可能机制。方法SD大鼠30只,随机分成对照组(Control组)、膝骨关节炎组(KOA组)和仙灵骨葆胶囊组(XLGB组),每组10只。HE染色观察软骨组织形态学变化;TUNEL法检测软骨组织细胞凋亡;ELISA法检测血清和关节液炎症因子水平;Western blot检测软骨组织金属蛋白酶、金属蛋白酶组织抑制因子和NF-κB信号通路蛋白表达。结果仙灵骨葆胶囊灌胃改善大鼠软骨组织形态,降低软骨组织细胞凋亡水平,抑制血清和关节液炎症因子IL-1β、IL-6和TNF-α水平,降低金属蛋白酶蛋白表达,增加金属蛋白酶组织抑制因子蛋白表达,抑制NF-κB信号通路。结论仙灵骨葆胶囊改善大鼠膝骨关节炎,其作用机制可能与NF-κB信号通路有关。 Objective To explore the therapeutic effect of Xian-ling-gu-bao capsule on rat knee osteoarthritis and its possible mechanism.Methods Thirty SD rats were randomly divided into control group,knee osteoarthritis group(KOA group)and Xian-ling-gu-bao capsule group(XLGB group),with 10 rats in each group.The histological changes of cartilage were observed by HE staining.TUNEL assay was used to detect the apoptosis of cartilage cells.The levels of inflammatory factors in serum and joint fluid were detected by ELISA.The expressions of metalloproteinase,metalloproteinase tissue inhibitor and NF-κB signaling pathway related protein in cartilage were detected by Western blot.Results Intragastric administration of Xian-ling-gu-bao capsule improved the morphology of cartilage tissue and decreased the apoptotic level of cartilage tissue cells.In terms of inflammation,the content of IL-1β,IL-6 and TNF-αin serum and articular fluid was reduced.Xian-ling-gu-bao capsule reduce the expression of metalloproteinase protein,increase the expression of tissue inhibitor of metalloproteinase protein and inhibit NF-κB signal pathway.Conclusion Xian-ling-gu-bao capsule can improve knee osteoarthritis in rats,and the mechanism may be related to NF-κB signaling pathway.
