摘要
目的:探讨儿童呼吸道上皮细胞中的干扰素λ1(IFN-λ1)在流感病毒(IFV)感染后的表达变化.方法:回顾分析了2017年10月~2019年1月在湖南省人民医院儿童中心住院的急性呼吸道感染性疾病的儿童鼻咽拭子标本,进行了11种儿童常见呼吸道病原核酸的检测,筛选出了100例A型流感病毒(IFVA)感染组(仅IFVA阳性),99例B型流感病毒(IFVB)感染组(仅IFVB阳性),92例呼吸道合胞病毒(RSV)感染组(仅RSV阳性),90例人鼻病毒(HRV)感染组(仅HRV阳性)儿童,56例健康体检儿童作为阴性对照组,均采用荧光定量PCR(qRT-PCR)方法对鼻咽拭子标本的IFN-λ1表达及病毒拷贝量进行检测.结果:在IFVA及IFVB感染组不同年龄组间IFN-λ1表达量无差异.IFVA及IFVB感染组IFN-λ1表达量分别较对照组升高,且均较RSV感染组及HRV感染组升高.IFN-λ1表达量在IFVA及IFVB感染组分别与其病毒拷贝量无明显相关,但与流感病毒感染后的临床评分存在正相关.结论:IFN-λ1可经IFV诱导在呼吸道上皮细胞表达,且与IFV感染儿童的疾病严重程度相关,推测IFN-λ1参与了机体抗IFV过程.
Objective To explore the expression change of interferonλ1(IFN-λ1)in respiratory epithelial cells of children after influenza virus(IFV)infection.Methods Nasopharyngeal swab specimens of children with acute respiratory infectious diseases who were admitted into Children’s Center of Hunan Provincial People’s Hospital from October 2017 to January 2019 were analyzed retrospectively.Nucleic acid detection of 11 common respiratory pathogens in children was carried out,and there were 100 cases in influenza vims A(IFVA)infection group(only positive IFVA),99 cases in influenza virus B(IFVB)infection group(only positive IFVB),92 cases in respiratory syncytial virus(RSV)infection group(only positive RSV),90 cases in human rhinovirus(HRV)infection group(only positive HRV)and 56 healthy children in negative control group.Real-time quantitative PCR(qRT-PCR)was used to detect the expression of IFN-λ1 in nasopharyngeal swab specimens in IFVA and IFVB infection groups.Results There was no difference in IFN-X1 expression among different age groups.The expression level of IFN-λ1 in IFVA and IFVB infection groups was higher than that in control group,and higher than that in RSV infection group and HRV infection group.The expression level of IFN-λ1 in IFVA and IFVB infection groups had no significant correlation with the vims copy numbers,but had a positive correlation with the clinical score after influenza virus infection.Conclusion After IFV infection,IFN-X1 can be expressed in respiratory epithelial cells,and it is related to the disease severity of children infected with IFV.It is speculated that IFN-λ1 is involved in the anti-IFV process.
