UPLC-Q-Exactive Orbitrap-MS法分析绵萆薢的化学成分 被引量:7

Analysis of Chemical Composition of Dioscorea spongiosa by UPLC-Q-Exactive Orbitrap-MS

摘要 目的采用超高效液相色谱串联四极杆-静电场轨道阱质谱(UPLC-Q-Exactive Orbitrap-MS)对绵萆薢的化学成分进行快速鉴别。方法采用Welch RP-C_(18)(150 mm×2.1 mm,1.8μm)色谱柱,以0.1%甲酸水溶液(A)-0.1%甲酸乙腈(B)为流动相进行梯度洗脱,体积流量0.30 mL·min^(-1),柱温35℃,分别以正负离子模式切换扫描,结合数据库匹配度、成分裂解规律与参考文献进行解谱和鉴别。结果共鉴定出绵萆薢化学成分30种,包括氨基酸与类氨基酸6种、有机酸类10种、黄酮类2种、甾体皂苷类1种、生物碱类2种、酰胺类1种及其他成分8种。结论该法可准确、快速地对绵萆薢中化学成分进行定性分析,可为绵萆薢的药效物质基础研究和质控指标的优选奠定实验基础。 Objective To rapidly analyse the chemical components of Dioscorea spongiosa by ultra-high performance liquid chromatography coupled with hybrid quadrupole-orbitrap mass spectrometry(UPLC-Q-Exactive Orbitrap-MS).Methods Welch RP-C_(18)(150 mm×2.1 mm 1.8μm)was used with gradient elution containing 0.1%formic acid aqueous solution(A)and 0.1%formic acid acetonitrile(B)at a flow rate of 0.30 mL·min^(-1) and a column temperature of 35℃.The spectrum resolutions were performed after scanned under positive and negative ion mode respectively,and then identified by database matching combined with composition cracking law and references.Results A total of 30 chemical components were identified from D.spongiosa,including 6 amino acids,10 organic acids,2 flavonoids,1 steroidal saponin,2 alkaloids,1 amide and 8 else components.Conclusion The method could be used for accurate and rapid qualitative analysis of D.spongiosa,and lay an experimental foundation for the basic study of the pharmacodynamic substances and the optimization of quality control indexes for D.spongiosa.
