摘要
为探究miR156过量表达对白桦扦插生根的影响,以白桦为材料,利用Gateway技术将包含miR156成熟序列的254 bp片段构建到p GWB2植物过表达载体上,采用农杆菌介导法进行遗传转化,获得8个miR156过表达白桦株系;对非转基因对照株系及转基因白桦株系进行嫩枝扦插,并通过石蜡切片对过表达株系扦插不定根发生过程进行解剖观察,分析miR156对扦插生根的影响。结果表明:miR156过表达白桦株系扦插生根率远高于野生型,其诱导根原基发生更早,说明转入miR156可以促进白桦嫩枝扦插不定根发生。
To explore the effect of miR156 overexpression on the rooting of Betula platyphylla cuttings, B.platyphylla was used as the material, and the 254 bp fragment containing the mature sequence of miR156 was constructed on the pGWB2 plant overexpression vector using Gateway technology. The genetic transformation was carried out by agrobacterium-mediated method, and 8 miR156 overexpression transgenic lines were obtained;softwood cutting was carried out on non-transgenic control line and transgenic lines, and the adventitious root formation process of over expressed lines was dissected and observed by paraffin section, so as to analyze the effect of miR156 on cutting rooting. The results showed that the adventitious rooting rate of B. platyphylla cuttings overexpressing miR156 was much higher than that of the wild type, and the induced root primordia occurred earlier, indicating that miR156 could promote the adventitious roots of B. platyphylla shoots.
