钙/钙调蛋白依赖性蛋白激酶Ⅱδ可变剪接与心脏疾病的关系

Relations between CaMKⅡδ alternative splicing and heart diseases

摘要 钙/钙调蛋白依赖性蛋白激酶Ⅱ(CaMKⅡ)是许多心脏疾病的关键酶,有4种亚型,其中CaMKⅡδ亚型发挥主要作用。由于CaMKⅡ表达和分布的复杂性,开发特异、有效的CaMKⅡ抑制剂一直是一个难题。RNA剪接被认为是调控心脏生长发育和疾病的关键因素。CaMKⅡδ在心脏主要存在4种剪接变体:δA、δB、δC、δ9,其对于心脏的发育和心脏疾病的发生发展作用各不相同。该文从基因结构、生物学特性、与心脏疾病关系等方面对CaMKⅡδ在心脏中4种剪接变体的研究进展进行总结,扩展对CaMKⅡδ在心脏生理和病理中作用的认识,为开发新的CaMKⅡ抑制剂提供思路。 The Ca;/calmodulin dependent protein kinaseⅡ(CaMKⅡ)is a key enzyme in many heart diseases.It has four subtypes,and CaMKⅡδsubtypes play a major role.Because of the complexity of CaMKⅡexpression and distribution,it has been a difficult problem to develop specific and effective CaMKⅡinhibitors.RNA splicing is considered to be a key factor in the regulation of cardiac growth and development and disease.At present,it is considered that there are four splicing variantsδA,δB,δC andδ9 of CaMKⅡδin the heart,which have different effects on the occurrence and development of the heart and heart disease.This paper summarizes the research progress of four splicing variants of CaMKⅡδin heart from the aspects of gene structure,biological characteristics and the relationship between CaMKⅡδand heart disease,expands the understanding of the role of CaMKⅡδin cardiac physiology and pathology,and provides ideas for the development of new CaMKⅡinhibitors.
