摘要
目的基于超高效液相色谱(UPLC)特征图谱,建立快速、有效鉴别南、北柴胡配方颗粒的方法。方法采用色谱柱ACQUITY UPLC®BEH C_(18)(2.1 mm×100 mm,1.7μm),流动相为乙腈-水,流速0.4 mL·min^(-1)梯度洗脱,检测波长为211 nm,250 nm,柱温35℃,对南、北柴胡配方颗粒进行特征图谱测定,分析特征峰差异。结果特征图谱方法精密度、稳定性良好,表明建立的方法合理、可行。其中北柴胡配方颗粒特征图谱有9个共有峰,南柴胡配方颗粒特征图谱有12个共有峰,南、北柴胡配方颗粒有7个共有峰,说明两者主要化学成分大体相同,个别成分存在一定差异。以特征峰差异可以对南、北柴胡配方颗粒进行区分,并进一步确定了南、北柴胡配方颗粒鉴别的直观区分点(β值)。结论UPLC特征图谱可对南、北柴胡配方颗粒进行质量评价,且能快速、准确、直观地反映南、北柴胡配方颗粒的基原差异,为南、北柴胡配方颗粒合理的临床应用提供重要的数据支撑。
OBJECTIVE To establish a fast and effective method for the identification of Bupleurum chinense formula granules and Bupleurum scorzonerifolium formula granules based on UPLC characteristic atlas.METHODS The ACQUITY UPLC®BEH C_(18)(2.1 mm×100 mm,1.7μm)chromatographic column was used,gradient elution was conducted with mobile phase consisting of acetonitrile and water,the flow rate was 0.4 mL·min^(-1),the detection wavelength was set at 211 nm and 250 nm,and the column temperature was maintained at 35℃.The characteristic chromatograms of Bupleurum chinense formula granules and Bupleurum scorzonerifolium formula granules were determined,and the difference of characteristic peaks was analyzed.RESULTS The precision and stability of the characteristic atlas method were good,which indicated that the method was reasonable and feasible.There were nine common peaks in the characteristic atlas of Bupleurum chinense formula granules,12 common peaks in the characteristic atlas of Bupleurum scorzonerifolium formula granules,and seven common peaks in the Bupleurum chinense and Bupleurum scorzonerifolium formula granules,which indicated that the main chemical compositions of the two ingredients were basically the same,but there were some differences in other components.According to the difference of characteristic peaks,the formula granules of Bupleurum chinense and Bupleurum scorzonerifolium could be distinguished,further the visual distinguishing point-βvalue of was determined.CONCLUSION The UPLC characteristic atlas can not only evaluate the quality of Bupleurum chinense and Bupleurum scorzonerifolium formula granules,but also quickly,accurately and intuitively reflect the differences of the basic elements between the Bupleurum chinense and Bupleurum scorzonerifolium formula granules,and provide important data support for the reasonable clinical application of the Bupleurum chinense and Bupleurum scorzonerifolium formula granules.
