Fscn_(2)基因过表达对顺铂诱导的耳蜗细胞凋亡的抑制作用

摘要 目的研究Fscn_(2)基因的过表达对顺铂诱导耳蜗细胞凋亡的影响。方法构建Fscn_(2)过表达的细胞模型,用30μM的顺铂干预Fscn_(2)过表达的细胞和对照组E.V.细胞24 h,CCK-8法检测细胞的增殖能力以及流式细胞术检测细胞凋亡率,Western Blot检测凋亡相关蛋白。结果在顺铂干预下,与对照组细胞相比,Fscn_(2)过表达的细胞增殖能力较强,凋亡细胞较少,抗凋亡蛋白Bcl-2的表达较高,凋亡相关蛋白Cleaved caspase3的表达较低。结论Fscn_(2)基因的过表达抑制了顺铂诱导的耳蜗细胞凋亡。 Objective To study the effects of Fscn_(2) gene overexpression on cisplatin-induced apoptosis of cochlear cells.Methods Cell model of Fscn_(2) overexpression was constructed,and 30μM cisplatin was used to intervene Fscn_(2) overexpressed cells and E.V.cells in the control group for 24 hours.Proliferation ability was detected by CCK-8 kit,the apoptosis rate was determined by flow cytometry,and the expression of apoptosis-related proteins was detected by Western blot.Results Compared with cells in the control group,Fscn_(2) overexpressed cells had stronger proliferation ability,fewer apoptotic cells,higher expression of anti-apoptotic protein Bcl-2,and lower expression of apoptotic protein Cleaved caspase3.Conclusion Overexpression of Fscn_(2) gene inhibits cisplatin-induced cochlear cell apoptosis.
