摘要
目的:探讨吸烟对DNA聚合酶polδ最小亚基POLD4 mRNA表达水平的影响。方法:于2018年01月至2018年05月收集我院无吸烟患者10例,轻度吸烟患者5例,中度吸烟患者8例,重度吸烟患者10例的血清各5 mL。标本经处理后通过RT-PCR检测polδ聚合酶各亚基POLD4、POLD1、POLD2、POLD3及DNA修复相关蛋白XPA、XPC的mRNA相对表达水平。结果:相对于无吸烟组,吸烟组POLD4 mRNA表达水平总体呈现下降趋势,尤其轻度吸烟组下降最明显,同为DNA聚合酶polδ催化亚基的POLD1及POLD3亚基的mRNA表达水平总体也呈下降趋势,但亚基POLD2 mRNA表达水平却呈反向增高。进一步对比同为DNA修复蛋白的XPA mRNA表达水平同POLD4类似吸烟组呈现出下降,尤其是轻度、中度吸烟组明显,而另一DNA修复蛋白XPC mRNA总体水平并未受吸烟影响,但亚组分析显示重度吸烟组表达水平下降。结论:吸烟可导致血清POLD4 mRNA表达水平下降,可能是导致肺癌形成的一个原因。
Objective:To investigate the effect of smoking on DNA polymerase polδsmallest subunits POLD4 mRNA expression level.Methods:From January 2018 to May 2018,serum samples of 5 mL were collected from 10 non-smoking patients,5 mild smoking patients,8 moderate smoking patients and 10 heavy smoking patients in our hospital.After treatment,the mRNA relative expression levels of POLD4,POLD1,POLD2,POLD3 and DNA repair related proteins XPA and XPC were detected by RT-PCR.Results:Compared to the non-smoking groups,in smoking group,POLD4 mRNA expression level showed an overall downward trend,especially in patients with mild smoking group,at the same time,DNA polymerase polδcatalytic subunit POLD1 and POLD3 mRNA expression level showed an overall downward trend,but the POLD2 mRNA expression level was increased.Further comparison showed that the expression level of XPA mRNA,which is also a DNA repair protein,was decreased in the smoking group,especially in the mild and moderate smoking group,while the overall expression level of another DNA repair protein XPC mRNA was not affected by smoking,but the subgroup analysis showed that the expression level of XPC mRNA was decreased in the heavy smoking group.Conclusion:Smoking can decrease the expression of serum POLD4 mRNA,which may be one of the causes of lung cancer.
