杞菊景明颗粒在大鼠血浆、胆汁、尿液、粪便中代谢产物的鉴定 被引量:3

摘要 目的鉴定杞菊景明颗粒在大鼠血浆、胆汁、尿液、粪便中的代谢产物。方法12只大鼠随机分为2组,分别灌胃给予生理盐水、颗粒浓缩液(5.46 g/kg),采集血浆、胆汁、尿液、粪便。UPLC-Q-TOF/MS分析采用ACQUITY UPLC BEH C_(18)色谱柱(2.1 mm×100 mm,1.7μm);流动相0.1%甲酸-乙腈,梯度洗脱;体积流量0.4 mL/min;柱温30℃;电喷雾离子源,正负离子模式。结果共鉴定出10种原型成分及20种相关代谢产物,其代谢途径包括Ⅰ相代谢反应(羟基化、去羟基化、还原)和Ⅱ相代谢反应(甲基化、葡萄糖醛酸化、硫酸酯化)。结论该方法稳定可靠,可为明确杞菊景明颗粒效应物质及其体内过程提供依据。 AIM To identify the metabolites of Qiju Jingming Granules in rat plasma,bile,urine and feces.METHODS Twelve rats were randomly assigned into two groups and given intragastric administration of normal saline and granule concentrated solution(5.46 g/kg),respectively,after which plasma,bile,urine and feces were collected.The UPLC-Q-TOF/MS analysis was performed on a 30 ℃ thermostatic ACQUITY UPLC BEH C_(18 )column(2.1 mm×100 mm,1.7 μm),with the mobile phase comprising of 0.1%formic acid-acetonitrile flowing at 0.4 mL/min in a gradient elution manner,and electrospray ionization source was adopted in positive and negative ion modes.RESULTS Total 10 prototype components and 20 metabolites were identified,whose metabolic pathways contained phaseⅠmetabolic reactions(hydroxylation,dehydroxylation,reduction) and phaseⅡmetabolic reactions(methylation,glucuronidation,sulfatation).CONCLUSION This stable and reliable method can lay a foundation for clarifying the effective substances and their in vivo processes of Qiju Jingming Granules.
