摘要
目的 研究清肺汤对急性呼吸窘迫综合征大鼠肺组织损伤和炎性介质释放的影响。方法 将SPF级SD雄性大鼠50只随机分为空白组、模型组及清肺汤高、中、低剂量组,每组10只。除空白组外均气管内滴注3 m/kg脂多糖(LPS)100μL,两肺均匀分布LPS 6 h,开始第1次给药;空白组、模型组给予灌胃蒸馏水10 mL/kg,清肺汤高、中、低剂量组分别灌胃清肺汤生药20、10、5 g/kg溶于10 m L/kg蒸馏水中;各组早晚各给药1次,连续3 d。干预结束后比较肺组织湿干重比(W/D);HE染色比较病理学形态变化;ELISA检测比较C反应蛋白(CRP)、降钙素原(PCT)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-10(IL-10)炎性介质释放情况。结果 与空白组比较,模型组W/D、CRP、PCT、TNF-α、IL-6、IL-10均升高,差异具有统计学意义(P <0.05);与模型组比较,清肺汤高、中、低剂量组W/D、CRP、PCT、TNF-α、IL-6、IL-10均下降,差异具有统计学意义(P <0.05);清肺汤高剂量组各项观察指标均优于中、低剂量组(P <0.05),HE染色可见清肺汤治疗后各组大鼠与模型组比较肺组织炎性渗出明显减轻。结论 清肺汤可能参与调节急性呼吸窘迫综合征大鼠炎症介质释放,减轻肺损伤。
Objective:To study the effect of Qingfei Decoction on lung tissue injury and inflammatory mediator release in rats with acute respiratory distress syndrome.Methods:Fifty SPF SD male rats were randomly divided into blank group,model group,high-dose Qingfei Decoction group,middle-dose Qingfei Decoction group,and lowdose Qingfei Decoction group,10 in each group.Except for the blank group,100 μL of 3 m/kg LPS was intratracheally instilled,and the LPS was evenly distributed in both lungs for 6 h,and the first administration was started;the blank group and the model group were given 10 mL/kg of distilled water;the high-,middle-and low-dose groups were respectively given Qingfei Decoction 20,10,and 5 g/kg dissolved in 10 mL/kg of distilled water;each group was administered once in the morning and evening for 3 consecutive days.After the intervention,the wet to dry weight ratio(W/D)of lung tissue was compared;HE staining was used to compare the pathological changes.Results:Compared with the blank group,W/D,CRP,PCT,TNF-α,IL-6 and IL-10 in the model group were all increased,and the difference was statistically significant(P < 0.05).The W/D,CRP,PCT,TNF-α,IL-6,IL-10 of Qingfei Decoction high-,middle and low-dose groups all decreased,and the difference was statistically significant(P <0.05);compared with the Qingfei Decoction middle-and low-dose groups,all the observation indexes in the highdose group were better(P < 0.05).HE staining showed that lung tissue inflammatory exudation was significantly reduced in each Qingfei Decoction group than the model group.Conclusion:Qingfei Decoction may be involved in regulating the release of inflammatory mediators and reducing lung injury in rats with acute respiratory distress syndrome.
