达氏鳇血细胞的组成、发生及吞噬功能研究 被引量:1

摘要 本文利用Wright’s染色技术,观察了平均体质量为(324.24±214.7)g的达氏鳇(Huso dauricus)外周血形态特征、血细胞发生;将调整浓度后的血液和酵母液加入灭菌后的离心管中充分混合30 s,30℃恒温水浴锅孵育40 min后涂片,干燥后用2.5%戊二醛固定6 min,Wright's染色、封片,进行血细胞吞噬实验,分析其免疫功能。结果表明,达氏鳇外周血细胞由红细胞、血栓细胞、淋巴细胞、单核细胞和粒细胞等组成;单核细胞体积最大,平均大小(长径×短径)为(13.10±1.56)μm×(11.10±2.45)μm,小淋巴细胞体积最小,平均大小(长径×短径)为(7.31±1.21)μm×(6.07±1.35)μm。印片结果显示,头肾,体肾和脾脏为达氏鳇的主要造血器官。红细胞,粒细胞和单核细胞主要在头肾和体肾中发生;淋巴细胞主要在脾脏发生,肝脏中未发现原始血细胞。酵母菌的吞噬实验表明,达氏鳇血细胞具有吞噬功能,红细胞的吞噬率为(22.84±3.21)%,粒细胞和单核细胞具有黏附能力,这与达氏鳇自身的免疫功能有关。 The morphological characteristics and hematopoiesis of the peripheral blood of huso sturgeon Huso dauricus with body weight of(324.24±214.7)g were observed using Wright's staining technique;the blood and yeast solution after adjusting the concentration were added to the sterilized centrifuge tubes were mixed thoroughly for 30 s,incubate in a constant temperature water bath at 30℃for 40 min,and then smear the smears.After drying,they were fixed with 2.5%glutaraldehyde for 6 min,stained with Wright's,mounted,and subjected to hemophagocytosis test to analyze their immune function.The results showed that the peripheral blood cells of huso sturgeon were composed of erythrocytes,thrombocytes,lymphocytes,monocytes and granulocytes,with the maximal volume and average size(long diameter×short diameter)of(13.10±1.56)μm×(11.10±2.45)μm in monocytes,and the minimal volume in lymphocytes,and the average size(long diameter×short diameter)of(7.31±1.21)μm×(6.07±1.35)μm.The blots showed that the main haematopoietic organs in huso sturgeon were head kidney,body kidney and spleen in which cytogenesis of erythrocytes,granulocytes and monocytes occurred mainly in the head kidney and body kidney,lymphocytes occurred mainly in the spleen,and no hematopoietic cells were found in the liver.Phagocytosis trial showed that blood cells of huso sturgeon had phagocytic function,with the phagocytosis rate of(22.84±3.21)%,and granulocytes and thrombocyte had adhesion ability,which are related to the immune ability of huso sturgeon.
