不同变型与产地天麻中6种多酚类成分的比较

Comparison of six polyphenols in Gastrodia elata from different variants and origin

目的比较不同变型和产地天麻中6种多酚类成分的含量差异。方法采用X Select^(■)HSS C_(18)色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈(A)-0.05%磷酸水溶液(B),梯度洗脱,检测波长220 nm,流速1 mL·min^(-1),柱温30℃,PDA检测器。结果6种成分回归曲线的r均>0.9995,精密度的RSD为0.31%~0.70%,稳定性的RSD为1.18%~2.79%,重复性的RSD为0.98%~2.41%;平均回收率为98.23%~101.74%,RSD为1.08%~2.39%。四川乐山产野生乌天麻中多酚的总含量最高(33.24±0.05 mg·g^(-1)),贵州德江产黄天麻最低(6.26±1.28 mg·g^(-1));在乌天麻中,四川乐山产总多酚的含量最高,湖北随县产的最低;在红天麻中,云南产总多酚的含量最高。结论所用方法准确、稳定,可有效测定不同变型及产地天麻中6种多酚类成分的含量。

OBJECTIVE To study the content differences of six polyphenols in Gastrodia elata from different varieties and origins.METHODS Chromatographic conditions include a X Select^(■)HSS C_(18) column(250 mm×4.6 mm,5μm)and the mobile phase consisting of acetonitrile(A)-0.05%phosphoric acid aqueous solution(B)with a gradient program.The flow rate was 1 mL·min^(-1) with the wavelength of 220 nm and the column temperature of 30℃.RESULTS The methodological linear correlation coefficients of the six components were all above 0.9995,with the precision RSDs of 0.31%-0.70%,the stability RSDs of 1.18%-2.79%,and the repeatability RSDs of 0.98%-2.41%.The average recovery rate was 98.23%-101.74%,with the RSDs of 1.08%-2.39%.The total polyphenol content of wild G.elata in Leshan,Sichuan was the highest 33.24±0.05 mg·g^(-1),and the lowest was 6.26±1.28 mg·g^(-1) in Dejiang,Guizhou.Among the G.elata f.glauca,its content was the highest in Leshan,Sichuan,and the lowest in Suixian,Hubei.Among the G.elata f.elata,the Yunnan producing area had the highest content.CONCLUSION This method is accurate and stable,and can effectively determine the content of six polyphenols in G.elata with different varieties and origins.
