ELOVL6在人卵巢浆液性腺癌组织中的表达及对卵巢癌细胞增殖、侵袭及凋亡能力的影响

Expression of ELOVL6 in human ovarian serous adenocarcinoma and its effect on the proliferation,invasion and apoptosis of ovarian cancer cells

摘要 目的探讨超长链脂肪酸延伸酶6(elongase of very long chain fatty acids 6,ELOVL6)在人卵巢浆液性腺癌组织中的表达,并研究ELOVL6对人卵巢癌细胞增殖、侵袭及凋亡的影响.方法选取2018年4月至2020年12月在青海省第五人民医院行卵巢浆液性腺癌术的80例患者,收集其癌组织,另收集20例因其他疾病行切除手术的正常卵巢和输卵管组织,通过免疫组化法检测患者卵巢组织中ELOVL6蛋白阳性表达.以人卵巢癌细胞SKOV3和OAW28作为对照组,以转染ELOVL6的卵巢癌细胞作为转染组,采用qRT-PCR验证对照组和转染组ELOVL6 mRNA表达水平;MTT实验和Transwell实验分别观察ELOVL6对人卵巢癌细胞增殖侵袭能力的影响,流式细胞仪检测ELOVL6转染后对卵巢癌细胞凋亡的影响.结果免疫组化结果显示,ELOVL6在正常卵巢和输卵管组织中90%以上呈阳性表达,而在低级别浆液性腺癌组织中阳性表达率为44.00%,在高级别浆液性腺癌组织中阳性表达率为54.55%,差异有统计学意义(P<0.05);qRT-PCR结果显示,与对照组卵巢癌细胞比较,转染组的SKOV3和OAW28卵巢癌细胞中ELOVL6 mRNA表达水平显著升高(P<0.05);MTT结果表明,SKOV3细胞和OAW28细胞分别在转染ELOVL6培养36 h和48 h后,其增殖能力较对照组有显著性下降(P<0.05);Transwell实验表明,转染组SKOV3细胞穿膜数少于对照组细胞穿膜数[(201±9)个vs(323±12)个],转染组OAW28细胞穿膜数少于对照组细胞穿膜数[(158±4)个vs(286±2)个],差异均有统计学意义(P<0.05);流式检测结果表明,ELOVL6高表达能够明显促进卵巢癌细胞的凋亡.结论ELOVL6在卵巢癌组织中呈弱阳性表达,在卵巢癌细胞中mRAN呈低表达,对卵巢癌细胞的增殖和侵袭起到抑制作用,能够促进卵巢癌细胞的凋亡. Objective To investigate the expression of elongase of very long chain fatty acids 6(ELOVL6) in human ovarian serous adenocarcinoma tissues, and to study the effect of ELOVL6 on the proliferation, invasion and apoptosis of human ovarian cancer cells.Methods A total of 80 patients who underwent ovarian serous adenocarcinoma surgery in the Fifth People’s Hospital of Qinghai Province from April 2018 to December 2020 were selected, and their cancer tissues were collected, and another 20 cases of normal ovarian and fallopian tube tissues who underwent resection for other diseases were collected.The positive expression of ELOVL6 protein in ovarian tissue of patients was detected by immunohistochemistry.Human ovarian cancer cells SKOV3 and OAW28 were used as the control group, and ovarian cancer cells transfected with ELOVL6 were used as the transfection group.qRT-PCR was used to verify the expression levels of ELOVL6 mRNA in the control group and the transfection group.MTT assay and Transwell assay were used to observe the effect of ELOVL6 on the proliferation and invasion ability of human ovarian cancer cells, respectively.The effect of ELOVL6 on apoptosis of ovarian cancer cells was detected by flow cytometry.Results Immunohistochemical results showed that ELOVL6 was positively expressed in more than 90% of normal ovarian and fallopian tube tissues, while the positive expression rate was 44.00% in low-grade serous adenocarcinoma tissues and 54.55% in high-grade serous adenocarcinoma tissues, the differences were statistically significant(P<0.05);The results of qRT-PCR showed that compared with the ovarian cancer cells of the control group, the expression levels of ELOVL6 mRNA in the SKOV3 and OAW28 ovarian cancer cells of the transfection group were significantly increased(P<0.05).After 36 hours and 48 hours of culture after transfection with ELOVL6 in the SKOV3 and OAW28 ovarian cancer cells, its proliferation ability was significantly decreased compared with the control group(P<0.05);Transwell experiments showed that the number of trausmembrane SKOV3 cells in the transfection group was less than the number of cells in the control group [(201±9) vs(323±12)],and the number of trausmembrane OAW28 cells in the transfection group was less than that in the control group [(158±4) vs(286±2)],the differences were statistically significant(P<0.05).The results of flow cytometry showed that the high expression of ELOVL6 could significantly promote the apoptosis of ovarian cancer cells.Conclusion ELOVL6 was weakly expressed in ovarian cancer tissues, and mRNA was lowly expressed in ovarian cancer cells, which inhibited the proliferation and invasion of ovarian cancer cells and promoted the apoptosis of ovarian cancer cells.
