摘要
目的观察COPD肺组织中MAL、IL-10、TNF-α的表达。方法收集2012-2015年在南华大学附属第一医院接受肺癌全肺切除术的患者病历,选择非肿瘤浸润的邻近肺组织。根据COPD诊断标准和患者资料,将标本分为肺功能正常组和COPD组的肺组织,每组5例。取SD大鼠16只,全部为雄性健康体,按照每组8只随机分为两组,一组为COPD模型组,另一组为正常组。采用HE染色法对人及实验大鼠的肺组织进行形态观测,IHC检测人及大鼠肺组织中MAL的表达及分布,ELISA检测大鼠肺组织中MAL、TNF-α、IL-10的蛋白水平。结果罹患COPD的人及大鼠肺组织中MAL蛋白表达水平低于正常组(P<0.05);与正常组相比,COPD大鼠肺组织中有大量炎症细胞浸润,MAL和IL-10表达明显减弱,TNF-α表达上调,差异均具统计学意义。结论MAL在COPD患者和大鼠肺组织中表达下调,IL-10在大鼠肺组织中表达下调,TNF-α在大鼠肺组织中表达上调,MAL与IL-10、TNF-α的表达水平显著相关。
Objective To observe the expression of MAL,IL-10,and TNF-αin the lung tissue of Chronic Obstructive Pulmonary Disease(COPD).Methods Patients who underwent pneumonectomy for lung cancer at the First Affiliated Hospital of South China University from 2012 to 2015 were collected,and non-tumor-infiltrating adjacent tissues were selected.According to the COPD diagnostic criteria and patient data,the specimens were divided into the normal lung function group and lung tissue of the COPD group,with 5 cases in each group.Meanwhile,Sixteen SD rats,all healthy males,were randomly divided into two groups according to 8 rats in each group,one group was the COPD model group,and the other was the normal group.Morphological observation of human and rat lung tissue was performed by HE staining.IHC was used to detect the expression and distribution of MAL in human and rat lung tissue,and ELISA was used to detect the protein levels of MAL,TNF-α,and IL-10 in rat lung tissue.Results The expression level of MAL protein in lung tissue of humans and rats with COPD was lower than that of the normal group(P<0.05).Compared with the normal group,a large number of inflammatory cells were infiltrated in the lung tissue of COPD rats,the expressions of MAL and IL-10 were significantly reduced,the expression of TNF-αwas up-regulated,and the differences were statistically significant.Conclusion The expression of MAL is down-regulated in COPD patients and rat lung tissue,the expression of IL-10 is down-regulated in rat lung tissue,and the expression of TNF-αis up-regulated in rat lung tissue.The expression levels of MAL and IL-10 and TNF-αare significantly related.
