摘要
目的探讨miR-372在非小细胞肺癌(NSCLC)组织中表达,以及升高A549细胞中miR-372表达对细胞增殖和侵袭力的影响。方法选取2018年3月至2021年3月在我院接受治疗的NSCLC患者111例,qRT-PCR术检测NSCLC和癌旁组织中miR-372表达,培养A549细胞,并分别M组、NC组和N组,分别转染miR-372模拟序列、阴性对照物及不作处理,qRT-PCR术检测细胞中miR-372表达,利用CCK-8实验、Annexin V-FITC/PI染色和Transwell实验分别检测细胞增殖、凋亡及侵袭力。结果NSCLC和癌旁组织中miR-372表达量分别为(0.31±0.10)和(1.02±0.11),差异有统计学意义(t=50.031,P<0.001);低分化、TNM分期Ⅲ~Ⅳ期和发生淋巴结转移的NSCLC组织中miR-372表达量明显低于中-高分化、TNM分期Ⅰ~Ⅱ期和未发生淋巴结(P<0.05);M组细胞中miR-372表达量高于NC组和N组(P<0.05),24、48、72和96h时细胞OD值低于NC组和N组(P<0.05),细胞凋亡率高于NC组和N组(P<0.05),侵袭细胞数低于NC组和N组(P<0.05)。结论miR-372在NSCLC组织中呈低表达,升高A549细胞中miR-372表达可抑制细胞增殖并加速细胞凋亡,降低细胞侵袭力。
Objective To investigate the expression of miR-372 in non-small cell lung cancer(NSCLC)tissues,and the effect of elevated miR-372 in A549 cells on cell proliferation and invasiveness.Methods A total of 111 patients with NSCLC who were treated in our hospital from March 2018 to March 2021 were selected.qRT-PCR was used to detect the expressions of miR-372 in NSCLC and adjacent tissues.The A549 cells were cultured and divided into the M group,NC group,and N group,transfected with miR-372 mimic sequence,negative control,and no treatment in the group,respectively.qRT-PCR was used to detect the expression of miR-372 in cells.CCK-8 experiment,Annexin V-FITC/PI staining,and Transwell experiment were used to detect cell proliferation,apoptosis,and invasiveness,respectively.Results The expression level of miR-372 in NSCLC and adjacent tissues was(0.31±0.10)and(1.02±0.11),respectively,and the difference was statistically significant(t=50.031,P<0.001).The expression levels of miR-372 in NSCLC tissues with poorly differentiated,TNM stagesⅢto IV and lymph node metastasis were significantly lower than those of moderate to well-differentiated,TNM stagesⅠtoⅡand no lymph nodes(P<0.05).The expression level of miR-372 in the cells in the M group was higher than those in the NC group and the N group(P<0.05),the OD values at 24,48,72,and 96 h were lower than those in the NC group and the N group(P<0.05),while the cell apoptosis rate was higher than the NC group and the N group(P<0.05),the number of invasive cells was lower than the NC group and the N group(P<0.05).Conclusion The expression of miR-372 is low in NSCLC tissues.Increasing the expression of miR-372 in A549 cells can inhibit cell proliferation,accelerate cell apoptosis,and reduce cell invasiveness.
