甜瓜白粉病和蔓枯病抗性种质资源的筛选及其抗性来源分析 被引量:1

摘要 为了筛选甜瓜抗病材料,对40份甜瓜种质资源分别进行白粉病和蔓枯病抗性鉴定并对筛选到的抗病材料进行抗性来源分析。结果表明:采用苗期人工接种方法共鉴定到11份甜瓜白粉病抗性材料,采用离体叶片接种方法共鉴定到8份蔓枯病抗性材料。通过对鉴定到的白粉病和蔓枯病抗性材料进行抗性来源分析发现,9份材料可用白粉病抗性基因分子标记引物me4em37扩增出目的条带,表明其可能含有与抗病甜瓜收集一号相同的白粉病抗性基因;6份材料可用白粉病抗性基因分子标记引物CMBR120_(-172)扩增出目的条带,表明其可能含有白粉病抗性基因Pm-2F;2份材料可用蔓枯病抗性基因分子标记引物CMCT505扩增出目的条带,表明其可能含有蔓枯病抗性基因Gsb-1;1份材料可用蔓枯病抗性基因分子标记引物CMAT170a扩增出目的条带,表明其可能含有蔓枯病抗性基因Gsb-4。该研究可为甜瓜白粉病和蔓枯病抗性遗传改良及其抗病杂交组合的配制提供理论依据。 In order to screen the resistant materials,40 melon germplasm resources were used for identification of the resistance to powdery mildew and gummy stem blight,and the genetic resistance origin of the identified resistant sources were further analyzed.The results showed that a total of 11 melon germplasm resources with resistance to powdery mildew were identified by the artificial inoculation method in the live seed-plants;a total of 8 germplasm resources with resistance to gummy stem blight were identified by the in vitro leaf inoculation method.Through analysis of the resistance source of the disease-resistant materials,we found that the target band could be amplified in 9 materials using the molecular marker primer of me4em37that linked to powdery mildew resistance gene,indicating that these materials may contain the same powdery mildew resistance gene as the disease-resistant melon of No.1 Shouji;the target band could be amplified in 6 materials using the molecular marker primer of CMBR120_(-172) that linked to powdery mildew resistance gene,indicating that these materials may contain the powdery mildew resistance gene Pm-2F;the target band could be amplified in 2 materials using the molecular marker primer of CMCT505 that linked to gummy stem blight resistance gene,indicating that these materials may contain the gummy stem blight resistance gene Gsb-1;the target band could be amplified in 1 material using the molecular marker primer of CMAT170a that linked to gummy stem blight resistance gene,indicating that it may contain the gummy stem blight resistance gene Gsb-4.This study can provide a theoretical basis for the genetic improvement of the resistance to powdery mildew and gummy stem blight and for the preparation of hybrid combination with disease resistance in melon.
