猪伪狂犬病病毒gE基因缺失株毒株的鉴定

摘要 通过细胞传代,获得了35代次的PRV_(SX)-gE基因缺失毒株。对不同代次的病毒进行TCID_(50)测定、无菌检验、支原体检验、外源病毒检验、特异性检验,并通过动物感染试验测定毒株对猪的致病性。结果表明:不同代次之间病毒滴度无显著差异,且无支原体和外源病毒污染。PRV_(SX)-gE基因缺失毒株(10^(8.0)TCID_(50)/mL)可致仔猪发病。用PRV_(SX)-gE基因缺失毒株制备的油乳剂疫苗免疫小鼠和仔猪,发现其对小鼠保护率达90%以上,对猪保护率达100%。试验表明,PRV_(SX)-gE基因缺失毒株具有良好的免疫原性。 The PRV_(SX)-gE gene deletion virus strain was reproduction on ST cells for 35 generations.TCID_(50)determination,sterility test,mycoplasma test,exogenous virus test and specificity test were carried out for each generation of virus.The pathogenicity of the virus was determined by animal infection test.The results showed that there were no significant differences in virus titer among different generations and no bacterial contamination,mycoplasma and exogenous virus contamination in each generation;PRV_(SX)-gE gene deletion virus(10^(8.0)TCID_(50)/mL)could cause disease in piglets;Mice and piglets were immunized with inactivated oil emulsion vaccine prepared by PRV_(SX)-gE gene deletion virus strain,and the protective rate was over 90%in mice and 100%in pigs.In conclusion,the PRV_(SX)-gE gene deletion virus strain had good immunogenicity.
