Caveolin-1和ER-α36相互作用促进胶质瘤细胞生长 被引量:1

摘要 背景与目的 脑胶质瘤是一种常见的神经系统恶性肿瘤,预后差,死亡率极高。窖蛋白-1(Caveolin-1,Cav-1)是一种膜蛋白,在多种肿瘤发生发展过程中发挥重要作用。ER-α36是一种主要位于细胞膜上的新型雌激素受体,在多种肿瘤组织和细胞中介导膜起始的快速雌激素信号通路,参与细胞的生长、分化等生理和病理过程。我们在前期研究中发现Cav-1与ER-α36在胶质瘤U87细胞中共表达。本研究旨在探讨Cav-1和ER-α36在胶质瘤组织和细胞中的相互作用及对胶质瘤细胞生长的影响。方法 应用免疫组织化学法分析胶质瘤组织中Cav-1表达以及Cav-1与ER-α36和GFAP的共表达情况。采用Western blot方法检测雌激素E2对Cav-1表达的影响。结果 Cav-1在高级别胶质瘤组织中过表达。Cav-1与星形胶质细胞标志物GFAP和雌激素受体ER-α36在胶质瘤细胞中共表达。进一步分析表明,低浓度雌激素E2可促进ER-α36和Cav-1的表达。结论 在恶性胶质瘤细胞中,Cav-1和ER-α36相互作用共同参与肿瘤细胞生长,为临床治疗胶质瘤提供新的潜在治疗靶点。
