‘台农17号’菠萝组培再生体系的建立 被引量:2

Establishment of Tissue Culture Regeneration System of Pineapple Cultivar’Tainong No.17’

摘要 为建立‘台农17号’菠萝组培再生体系,以‘台农17号’菠萝增殖单芽为试验材料,研究不同激素处理对愈伤组织诱导、增殖、再生和不定芽增殖及生根的影响。结果表明暗培养条件下,愈伤组织诱导效果最好的培养基是MS+6-BA 3.0 mg/L+NAA 2.0 mg/L,4周内愈伤组织诱导率高达92%,愈伤组织重量达1.49 g;在此基础上分别添加2,4-D、KT、ZT后,效果最好的是添加2,4-D浓度0.20 mg/L处理,愈伤组织长势明显提升,重量值达到最高的1.73 g,而添加KT和ZT愈伤组织增殖效果和长势未得到显著提升;光照培养条件下,愈伤组织再生最佳配方是MS+6-BA 2.0 mg/L+NAA 2.0 mg/L,再生率达到82%;MS+6-BA 2.0 mg/L+NAA3.0 mg/L适宜不定芽增殖,4周后增殖系数达5.40;生根配方1/2MS+IBA 0.5 mg/L+NAA 0.50 mg/L生根效果最佳,平均生根条数约3.87条,生根率95%。研究明确了不同激素及配比对‘台农17号’菠萝愈伤组织诱导、增殖、再生和芽增殖及生根的影响,初步建立了‘台农17号’菠萝高效组培再生技术体系,为深入研究菠萝开花机理提供了良好基础。 In order to establish an effective tissue culture regeneration system for pineapple cultivar’Tainong No.17’,taking the proliferated single bud as experimental material,this paper studied the effects of different hormone treatments on the callus induction,callus propagation,callus regeneration,adventitious buds propagation and the rooting of pineapple cultivar’Tainong No.17’.The results showed that the medium of MS+6-BA 3.0 mg/L+NAA2.0 mg/L maked the highest callus induction rate of 92%and the heaviest weight of 1.49 g;Comparing the effects of 2,4-D,KT and ZT supplementation,the best treatment was adding 2,4-D at a concentration of 0.20 mg/L,and the callus growth was significantly improved,the weight value reached the highest of 1.73 g,while the callus proliferation and growth were not significantly improved by adding KT and ZT.The optimal medium for callus regeneration with regeneration rate of 82%under light was MS+6-BA 2.0 mg/L+NAA 2.0 mg/L,and the regeneration rate reached 82%;The medium of MS+6-BA 2.0 mg/L+NAA 3.0 mg/L was suitable for adventitious bud proliferation,and the proliferation coefficient reached 5.40 after 4 weeks.Rooting medium of 1/2MS+IBA0.5 mg/L+NAA 0.50 mg/L had the best rooting effect,the average number of roots was 3.87,and the rooting rate was 95%.The effects of different hormones and proportions on callus induction,proliferation,regeneration,bud proliferation and rooting of pineapple cultivar’Tainong No.17’were clarified in this study,and a highly efficient tissue culture and regeneration system was prelimatively established,which laid a good foundation for further research on the flowering mechanism of pineapple.
